Object:The nonsyndromic cleft lip and/or cleft palate (NSCLP)belongs to polygenic inheritance diseases and its nosogenesis still remains mistiness. The study explored the association of CTGF,TGFβ3 and their receptors (TGFβRⅡ,LRP6) with the generation of NSCLP.Method: There are 40 samples in experimental group and 18 samples in control group. The expressions of CTGF,TGFβ3 and their receptors (TGFβRⅡ,LRP6) in fissure tissues of the patients with NSCLP were measured by the SP immunohistochemistry and analyzed if there were some variabilites compared with unimpaired child, and further to analyze the variabilities of the expressions of genes above-mentioned among the different types/variant degree groups in experimental group as well as the variant age groups in experimental group.Results: (1) there was no variability for the expression of CTGF between the experimental group and the control group (P>0.05); there were no variabilities among the different types/ variant degree groups in experimental group as well as the variant age groups in experimental group (p>0.05). (2) The expression of CTGF receptor (LRP6) decreased in experimental group compared with that in control group (P<0.01), but had no difference among the different types/variant degree groups in experimental group as well as the variant age groups in experimental group (P>0.05). (3) Significant increase for the expression of TGFβ3 was observed in experimental group (P<0.01) but no significant difference was shown among the different types/variant degree groups in experimental group. And there were variables for the expression of TGFβ3 among the variant age groups in experimental group (P<0.01), older the age was, stronger the expression of TGFβ3 was. (4) there was no difference for the expression of TGFβ3 receptor (TGFβRⅡ) between the experimental group and the control group (p>0.05); There were no variabilities among the different types/ variant degree groups in experimental group as well as the variant age groups in experimental group (p>0.05).Conclusions: (1) The result gave no evidence for the association of CTGF with the generation of NSCL/P. (2) The result which the expression of CTGF receptor (LRP6) decreased in experimental group suggested that it had relationship with the generation of NSCLP. (3) The result supported that there was association between TGFβ3 and the generation of NSCLP and no evidence showed the expression of TGFβ3 had any relationship with deformed type and degree. (4) The research showed that the age was older, the expression of TGFβ3 was stronger in the patients and this could be one of reasonable explanations for the phenomenon that operative incisions usually take place more severe scar contracture in the younger patients with cleft lip than that in the elder. (5) The study did not support the association of TGFβRⅡwith the generation of NSCLP.
