BackgroundLiver fibrosis is to point to by all kinds of pathogenic factor connective tissue inside liver caused by abnormal growths, lead to liver inside diffuse the extracellular matrix of the pathology of excessive deposition process, and its development is the end result of cirrhosis of the liver. Studies have shown that, in the liver fibrosis development to cirrhosis of the liver process, Intestinal endotoxemia(IETM) plays an important role in promoting. Tutor Liu Lixin’s previous findings, Thioacetamide (TAA) in rat liver caused by injury of the simultaneous occurrence of IETM, its induced liver injury animal model and human liver injury characteristics consistent with, can better reflect the actual situation of human liver injury. According to reports in the literature, probiotics Bicifo (normally living bacterium joint capsule) can adjust the Intestinal flora, support anaerobic bacteria growth and reduce harmful bacterium proportion, so as to reduce lumen through the generation of endotoxin, has been widely applied in clinic.The data makes clear,probiotics bacillus licheniformis (bacillus licheniformis are living bacterium capsule) is a facultative anaerobic bacteria,aerobic condition can rapidly consume environment of free oxygen, have a strong biological capture oxygen effect, promote the growth of anaerobic bacteria, the metabolites produced during antibiotic active substance,and mainly used in the clinical treatment of patients with diarrhea, but its in improving the research level of liver fibrosis are rarely reported.In the research, probiotics Bifico and Bacillus licheniformis are supplied to the hepatic fibrosis rats, and we observe the effect Bifico and Bacillus licheniformis on hepatic fibrosis, focusing on the degree of liver fibrosis effect of Bacillus licheniformis on.ObjectiveTo investigate whether Bacillus licheniformis (Bacillus licheniformis are living bacterium capsule) can improve liver fibrosis degree through the improvement of Intestinal endotoxemia(IETM).Methods50males SD rats randomly divided into normal control group, TAA model group, Bifico group, the low-dose group of Bacillus licheniformis and the high-dose group of Bacillus licheniformis. Liver fibrosis is induced by TAA, HE staining is used to observe liver damage situation and Picric Sirius red staining to observe collagen deposition. Plasma endotoxin level, serum Alanine aminotransferase(ALT),Aspartate aminotransferase(AST), Lactate dehydrogenase(LDH) and Malondialdehyde(MDA) level are simultaneously tested.The correlation between liver tissue collagen fiber area percentage (Picric Sirius red staining) and plasma endotoxin level is analysed.Results(1) Plasma endotoxin level:The plasma endotoxin level of TAA model group rats is significantly higher than the normal control group (P<0.01). The plasma endotoxin level of high and low doses groups of Bacillus licheniformis and Bicifo group rats is significantly lower than TAA model group (P<0.05).(2) Serum ALT, AST, LDH levels:The serum ALT、AST、LDH levels of TAA model group rats are significantly higher than the normal control group (P<0.05).The serum ALT、 AST、LDH levels of Bicifo group rats are significantly lower than TAA model group (P<0.05), The serum AST、LDH levels of high and low doses groups of Bacillus licheniformis rats are significantly lower than TAA model group (P<0.05),But the serum ALT level doesn’t reduce (P>0.05)(3) Serum MDA level:The serum MDA level of TAA model group rats are significantly higher than the normal control group (P<0.05).The serum MDA level of Bicifo group、high and low doses groups of Bacillus licheniformis rats are significantly lower than TAA model group (P <0.05).(4) HE staining:Compared with the normal control group, the TAA model group rats was normal structure damage, liver cells arranged in disorder, around the central veins visible patchy necrosis and massive infiltration of inflammatory cells, and a fibrous septum formation, significant liver fibrosis formation (P<0.01); Compared with the TAA model group, high and low doses groups of Bacillus licheniformis and Bicifo group rats hepatic lobules of normal structure of liver cells, with a small amount of necrosis, inflammatory cell infiltration was less, the liver fibrosis degree were significantly improved (P<0.05).(5)Picric Sirius red staining:compared with the normal control group, the TAA model group rats in liver tissues of with a large red collagen fibers, and the pseudolobuli formation, significant liver fibrosis formation(P<0.05);Compared with the TAA model group, high and low doses groups of Bacillus licheniformis and Bicifo group rats liver tissue central venous tube wall around to see more red collagen fibers(P<0.05). Liver tissue collagen fiber area percentage and plasma endotoxin level have positive correlation(r=0.442,P<0.01).ConclusionThe Bacillus licheniformis can improve liver fibrosis degree through the improvement of IETM.
