Backgroung Bronchial asthma (asthma) is regarded as polygenic inheritable disease, and an interaction between susceptibility genes and environmental factors play an important role in the occurrence and development of asthma. More and more researchers to study the genetic causes of asthma. In recent years, the association research is one of the hot spot between the SNP of ADAM33gene and asthma. The results were different in the various race. At present, related research results had not been reported of the SNPs in ADAM33and asthma in Zhuang people.Objective Zhuang is one of the minorities in China, mainly live in Guangxi region. It is a significant difference in different races between different genetic background. So, the aim of this study was to evaluate whether SNPs T1and V4in ADAM33gene are associated with the asthma in a Zhuang population in Guangxi. Provided the genetic data in zhuang people.The research results will be the reference for further study about mechanism of ADAM33gene leading to asthma in the future.Methords Using case-control study method to finish. We recruited70 asthma patients in Asthma specialist out-patient in the First Affiliated Hospital of Guangxi Medical University and control group were from89unrelated healthy volunteers. Genomic DNA was extracted from peripheral venous blood. Method polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was use to genotype.To further verification, we randomly selected different genotypes PCR products to direct sequencing.Rrsults (1) The T1locus has been found to be A'G polymorphism.In78patients, the frequencies of genotype of A/A、G/G、and A/G were81.4%、0%、18.6%.And the frequencies of control group wree61.8%、2.2%、36%(P<0.05).The frequency of A and G alleles was90.7%and9.3%in asthma patients. They were79.8%and20.2%in control group (P<0.05).(2) There was polymorphism in V4locus (C'G).The genotype frequence of V4Locus were C/C14.3%、G/G31.4%、C/G54.3%in case,and they were C/C12.4%、G/G23.6%、C/G57.3%in controls.The allele frequence were C41.4%、G58.6%in case and C41%、G59%in controls. There were not significant differences in both genotype and allele in this locus.Conclusion (1) T1and V4locus were found to have polymorphisms in Guangxi zhuang population.(2)ADAM33gene polymorphism T1have relevant association with asthma in Guangxi zhuang population.(3) In Guangxi Zhunag population, ADAM33gene V4locus exist C'G polymorphism,but may have not association with the occurrence of asthma.
