Objective:1、Research and analysis the expression of tumor-suppressor gene P27in colorectal cancer(CRC), colorectal adenomas, colorectal hyperplasia polyps and normal colorectal mucosa, investigate the possibility of P27as an early diagnostic marker of CRC.2、Analysis the relationship between the expression of P27and Clinical and pathological features of CRC, investigate the significance of P27in the incidence,development and prognosis of CRC.Methods:Collect specimens of a total of180cases in Ningxia Medical University Affiliated Hospital from January12006to December312010, Including120cases of surgical resection specimens of colorectal cancer patients,20cases of biopsies specimens determined as colorectal adenomas and20cases determined as colorectal hyperplasia polyps reported by outpatient gastroscopy room as pathology cases,20cases determined as normal mucosa specimens by HE staining of department of pathology. Treated specimens using immunohistochemical method, Research and analysis of P27’s expression in CRC, colorectal adenomas, colorectal hyperplasia polyps and normal colorectal mucosa.Results:1. P27’s positive expression rate was48.33%(58/120) in CRC,95.50%(19/20) in colorectal adenomas,90.00%(18/20) in colorectal hyperplasia polyps, and85.00%(17/20) in normal colorectal mucosa. P27’s positive expression rate in colorectal cancer is lower than the rates in colorectal adenomas, colorectal hyperplastic polyps and normal mucosa(P<0.05). There was no significant difference of P27’s positive expression rate in pairwise comparisons in colorectal adenomas, colorectal hyperplastic polyps, and normal mucosa (P>0.003).2.The positive expression rate of P27in CRC is negatively correlated to the degree of differentiation of colorectal cancer, lymph node metastasis, distant metastasis and Dukes stage(P<0.05);and is irrelated to age, sex, tumor location(P>0.05).Conclusion1、The positive expression rate of P27in CRC is less than the rate in colorectal adenomas, colorectal hyperplastic polyps and normal mucosa, While there is no significant difference of P27’s expression in these three samples,which points out that P27may be involved in the incidence of CRC.2、P27’s expression is negatively correlated to the degree of differentiation of colorectal cancer, lymph node metastasis, distant metastasis And Dukes stage, consider the expression of P27may become one of the indicators to evaluate the prognosis of CRC.
