To examine the cell proliferation the cell line was treated by NS-398fordifferent densities and hours by MTT. Flow cytometric assay was used to detect the effect ofthe Tca8113treated by different densities of NS-398on cell apoptosis for24hours.Immunofluorescence was used to detect the MMP-2expression of the Tca8113treated by0、20、40、80μmol/L NS-398for24hours. Westrenblot was used to detect the MMP-2expression of the Tca8113treated by20、40、80μmol/L NS-398for24hours.Results:1.20、40、80μmol/L NS-398inhibit the proliferation of Tca8113cell in a noseand time dependent manners.2.The cell apoptosis was enhanced significantly when the Tca8113cell was treated withNS-398(20、40、80μmol/L) for24hours.3. Immunohistochemistry and westernblot testing showed that the expression of MMP-2wassignificantly decreased the NS-398(20、40、80μmol/L)treatment24hours group comparedwith the control group. And the high concentration group Comparison with the lowersconcentration greatly inhibited the expression of MMP-2. Conclusion:1. NS-398can inhibit cell growth and induce apoptosis on Tca8113cells significantly.2. NS-398can down-regulating expression of MMP-2on Tca8113cells.
