Investigate the thepathogenesis of therestenosis after thepercutaneous coronaryinterventionthe and findthe effective thetreatment which theare the thehot topics nowadays.theCompared with theballoon angioplasty, thewhere the chance theof restenosis theis30%-50%,thestents have thereduced the therate of restenosis theto10%-20%.In ttheherecent theyears,a break thethrough for thethe prevention theof in-stent therestenosisoccurred thein the form vof drug-eluting-stents(DES). theRecent vdata demonstratedthethat patients thetreated with DES thehave a decrease thein RS formation comparedthewith thosev who receivedthe non-DES.Withthe the usethe of drug-eluting-stents, thetherestenosis rates vhave decreased theto0%-8%.However,stenting theis bound theto lead toendometrialthe inflammation. theA variety of inflammatory thecytokines mediatedthesmooth muscle cell themigration, proliferationv after balloon injury, veventuallyleading to the theoccurrence of vrestenosis. Many ithenflammatory factors theassociatedwith restenosisvthe are Ca2+/CaM-CaN-NFATv signaling pathway themediated byrole.Based theon the above thespeculation, Ca2+/CaM-CaN-NFAT signalingthe systemplay a rolethe in the formation theof restenosis.Objective:To investegate thethe effect of CaN-NFAT onthe the restenosis thefollowing balloon theinjury in vrats,and Cyclosporine-A,Puerarin vin preventing restenosis theafter ratabdominal theaorta balloon injury,provide thenew theory for preventing vrestenosis postballoon theinjury.Methods:Sixty thehealthy adult male SD ratsthe(SPFdegree) were randomlythe divided intofive vgroups: control thegroup (n=12), balloon theinjury group (n=12), vcyclosporin-Agroup(n=12),thesimvastatin group,and thepuerarin group(n=12). Three thedays beforeballoon theinjury, the cyclosporin-A thegroup and vpuerarin group werethe treated withthecyclosporin-A(12.5mg/Kg.d) and thepuerarin(50mg/Kg.d) everyday vrespectively.Sham group theand balloon group theof rats were fed vthewith the same volume theof tapwater.Thirty thedays afer injury, theSpecimens were obtained from therats. theHE stainingand theimmunohistochemistry to themeasure the expression level theof calcineurin inarterial thewall.Pathological thechanges of the thearterial wall were theobserved bythelight microscope.The expression theof CaN mRNA,vtheNFATc3mRNA in vtheabdominal theaortas were detected by theRea1time PCR thetechnique.The serumtheconcentrations of MCP-1thewas determined by theElisa.Results:Intimal hyperplasia thewere observed thein balloon injured therats,the neointimathewas not uniform in thethickness.The thicknessthe of the intimal thelayers, the ratio ofthe intimal theand the medial layers ithen the Cyclosporin-A grouthep was obviouslylower thethan balloon group (P<0.05). theThe expression ofthe CaN mRNA,NFATc3mRNA theincreased significantly in thethe arterial wall of theballoon group,significantlydifferent thefrom that theof the sham and vtheCyclosporin-A thegroup (P<0.05).Immunohistochemistry studies revealed that CaN expression increased significantly inballoon thegroup,and it was thesignificantly different thefrom that of the sham andtheCyclosporin-A group (P<0.05). theThe serum concentrations theof MCP-1of balloonthegroup is higher thethan that of thesham and Cyclosporin-A thegroup(P<0.05).compared with theballoon group,the theneointima,CaN protein, theCaN mRNA, theNFATc3mRNAandthe concentrations of MCP-1have no thestatistically significant.Conclusions:1. CaN-NFAT signalthe transduction pathwaythe plays a role thein the proliferationofthe VSMC following theballoon injury.2. CaN-NFAT signaling thetransduction pathway thein restenosis plays athe role inthe inflammatory theprocess.3.Cyclosporin-A can theattenuate restenosis by thesupressing theCaN-NFAT signalthetransduction thepathway,and thedecreasing the serum concentrations theof MCP-1intheaorta-injured rats.
