Antagonism, esterase isozyme electrophoresis, ITS and IGS-2 analyses, The growth rate of hyphae, combing with cultivation of the hyphae and enzyme activity test, were employed on the taxonomic identification and genetic diversity of 62 cultivated Pleurotus spp. strains. The results are as follows:1. The ribosome ITS of the 62 strains were amplified with the primer of ITS1 and ITS4, and about 700bp segments were obtained. The results of sequencing and blasting experiments showed that 26 strains are Pleurotus pulmonarius, 24 strains are Pleurotus cornucopiae, 5 strains are Pleurotus citrinopileatus and 4 strains are Pleurotus sapidus, and 3strains are Pleurotus abalones.2. According to antagonism reaction, the stain 1120, the stain 1121and the stain 1122 in the same vegetative compatibility groups; like 1048 and 1049.3. The IGS-2 and The esterase isozyme electrophoresis analysis showed that strains are different and of ample polymorphism .4. According to The growth rate of hyphae, cultivation of the hyphae and enzyme activity test, the strain which is temperature sensitive has a higher activity atlaccase; or it has a higher activity at CMC.It was summarized that there is obvious discrepancy among the tested strains through Cluster Analysis using esterase isozyme, IGS-2 and ITS method, which indicated that there is rich genetic diversity of oyster mushroom strains. |
