In this text, the seed of Ligusticum sinensise Oliv. were selected as the trial material. Through the experiments such as seed physical properties, water permeability of seed coats, development of embryos and the biotic measurement of the methanol extract of the seeds, GA3 and stratification treatments to find out the reason of seed dormancy and how to break the dormancy. Through the changes of embryo and content of main storage substances during the stratification period to discuss the relation of the dormancy. The text in order to find out the methods that could break dormancy and develop artificial cultivation.(1)Through to anatomy observation of the seeds of Ligusticum sinensise Oliv. it showed that:The seed’germination rate is 16%, grain weight is 1.58g.The length and width of the seed is 4mm and 2-2.5mm.(2)Through to anatomy observation of the seeds of Ligusticum sinensise Oliv. it showed that:The seed had a tiny embryo.It was not fully differentiated when harvested, only occupied 9.3% of the seed, some of the seeds even don’t have embryo.the seeds of Ligusticum sinensise Oliv. are made up of seed capsule,embryo,endosperm.The proportion of three parts is 4.52%,2.03%,93.45%.This is the main reason of the dormancy. The extract of the seeds can inhibited the germination and the root length of the Chinese cabbage seeds, and the higher of the concentration, the stronger of the inhibitory effect. This is the second reason of the dormancy.(3)Dealing seed with proper concentration of GA3 can improve the germination rate. When the concentration of GA3 is 500mg/L, the immersing time is 24h, the germination rate can reach 63.33%.But when the concentration of GA3 is 1000mg/L, the germination rate begins to drop.(4)The study showed that:The cool temperature (4℃) stratification is the best method to break the dormancy, the germination rate can reach 90.33%,it is the highest of the three stratification temperature, The second temperature is 15~4℃. The germination rate of the warm temperature (15℃) stratification is the lowest 62%, it has the highest decay rate 64%. (5)The embryo of the seed continued to grow during the cool temperature (4℃) stratification, from 9.3% to 85.81%. In the course of the cold stratification, the transformation between the protein, and sugar could provide big energy for the sprouting of the seeds.(6)The germination rate under five temperature(10℃、15℃、20℃、25℃、30℃)is different.The best germination temperature is:25℃,The germination rate is 95.33%.The most appropriate germination temperature renge is:20~25℃.The light has no influence of the seeds.In a word, the reason of the Ligusticum sinensise Oliv. seeds dormancy is embryo morphology dormancy and physiology dormancy. The cool temperature (4℃) stratification is the best method to break the dormancy.
