Eascaphis konlshli Hayata belongs to theaceae and Euscaphis genera, it is unique native ornamental species of China. Its root or root bark、flowers and dry fruit are all used for medicine, its charaters are acrid、sweet and placid, its root can dispel rheumati-sm, strengthen spleen and treat dysentery、diarrhea、colic、rheumatic pain and traumatic injury. Flowers can be used for analgesia and treat headache and vertigo; dry fruit has an efficacy of regulating vital energy and detumescence, and can treat stomachache、hernia、diarrhea and prolapse of rectum, especially for people who is allergic for paint, it has a unique effect and it is a kind of good Chinese medicinal materials. In China, a lot of Chinese patent medicines, pharmacy companies are all attempt to use its fruit for pharmacy and dispensing, and try to set up tree base for medica, but because of lacking germchit, the progress is slowly. These was no report of seeding and cutting propagation until 2007, but till now there has not been a successful report about seedling of tissue culture. The research is based on studying team accomplish the study of biological characteristics and seedling breeding techniques of E. konlshli. In case of that it has a low germination rate of using conventional plantings and is difficult for popularizing the techniques of rapid multiplication of superior provenance, the research respectively use seed、stem and leaves of initial culture of E. konlshli as test materials for tissue culture experiment. The results showed that:1、The trial use the seed、stem and leaf of initial culture of E. konlshli for the tissue culture. The results show that the optimum subculture medium of seed differentiation MS+BA1.0 mg·L-1+ NAA0.5 mg·L-1+agar7.5 g·L-1+sugar20 g·L-1; The optimum subculture medium of bundle pullulatein duced and growth is BA2.0 mg·L-1+ NAA0.3 mg·L-1+IAA1.0 mg·L-1+agar7.5 g·L-1+sugar20 g-L-1; 1/2MS+NAA0.1 mg·L-1+IBA0.5 mg·L-1+agar7.5 g·L-1+sugar20 g·L-1 is the optimum rooting medium, the rooting rate is 95% and mean root length is 3.51 cm; 2、The optimum disinfection of stem reproduction is 75% alcohol 5 s+0.1% sublimate; MS+ZE2.0 mg·L-1+NAA0.5 mg·L-1+IAA1.0 mg·L-1+agar 7.5 g·L-1+sugar20 g·L-1 is the optimum subculture medium of stem growth;3、The optimum rooting medium.1/2MS+NAA 0.1 mg-L-1+IBA0.5 mg·L-1+agar7.5 g·L-1+sugar20 g·L-1; The optimum subculture medium of shoot prolifera tion and rooting are separately WPM+BA2.0 mg·L-+NAA0.3 mg·L-1+agar7.5 g·L-1+sugar20 g·L-1 and 1/2WPM+IBA0.5 mg·L-1+NAA0.1 mg·L-1+agar7.5 g·L-1+sugar20 g·L-1.
