Abstract/Summary:
Melastoma intermedium Linn. is a evergreen shrub, which belongs to MelastomaGene(Melastomataceae). It grows in Taiwan, Fujian, Guangdong, Hainan, Guangxi andGuizhou provinces and etal. It mainly located in Zhangzhou, Fuzhou and Nanping city inFujian province. It's an ideal urban greening shrub for it's colorful flowers. The plant containstannins, flavonoids, amino acids and other compounds with antioxidant, antimicrobial andother medicinal value, so it has great potential for development. This research based on thedistribution of resources and living conditions of the investigation, through the research of thetissue culture of the Melastoma intermedium to building the establishment of plant rapidpropagation and regeneration technology system, for improving the proliferation factor,researching the regeneration principle, meeting the needs of industrial production. The mainresults were described as follows:1. Melastoma intermedium germplasm resources of wild state distribution.The distribution of the Melastoma intermedium wild plants resources are mainly concentratedin the province of east, south and central of Fujian, vertical distribution at an altitudeof400-800m slopes or understory, the field survey found on the Melastoma intermedium ofFujian, their populations have dramatically reduced the distribution to shrink year by year,the living conditions can not be optimistic.2. The establishment of plant rapid propagation of Melastoma intermedium. Melastomaintermedium stem segment explants soaked in75%alcohol for30s, wash three times withsterile water, soaked in5drops0.1%HgCl2and tween for6min, wash five times with sterilewater, control pollution below25.33%. The best start medium was MS+6-BA1.0mg.L-1+NAA0.5-1.0mg.L-1, the germinating rate could reach67.57%, new seedling height1.72cm.The ideal subculture proliferation medium was MS+KT0.1mg.L-1+6-BA1.0mg.L-1+NAA0.5mg.L-1, after30days, multiplication coefficient could reach3.412, the best subculturemultiplication cycle was45days. The best medium for rooting was1/2MS+IBA0.1mg.L-1+NAA0.1mg.L-1, after30days, the best rooting rate could reach84.7%. The length ofroots could reach3.97cm, the quantity of roots could reach5.92. The seedlings transplantingof tissue culture in the medium of red soil, sand by2:1was best and the survival rate could reach68.0%.3. The regeneration system of Melastoma intermedium. Soaked the leaf explants ofMelastoma intermedium30s with75%alcohol, wash three times with Sterile water, soaked in0.1%HgCl2and tween for6min, wash five times with sterile water, control pollution below22.76%. The leaves callus induction rate was higher than stem segment, reaching84%. Thebest explant was the leaves on the back side up flat on the medium. The ideal medium forcallus induction was MS+2,4-D2.0mg.L-1+6-BA1.0mg.L-1, after30days, induction ratecould reach51.85%. To determine by comparing the size of the callus proliferation, the bestmedium for callus proliferation was MS+2,4-D0.1mg.L-1+6-BA2.0mg.L-1+NAA0.1mg.L-1. The best medium for callus redifferentiation was MS+6-BA1.0mg.L-1+NAA0.5mg.L-1.More than research works, which certainly have the important theoretical and realmeaning for protecting Melastoma intermedium resource, the sustainable use of resources andseeking effective technical support.
