Research On The Preventive And Therapeutic Effect Of Complex Prescription Kuqin On CPV And Pharmacodynamic Action

Canine parvovirus was first confirmed by the United States on 1978,and was quickly spreaded arround the world within 2 years.It is easy that erupt all the dogs in all the seasons to canine parvovirus,which induce very high death rate.So far,we have not a very efficient way to cure the disease,According to theory of traditional Chinese medicine,canine parvovirus is determined to dampness and heat diarrhea,which is dysfunction of the spleen in transporting and distributing food essence and dampness,due to dampness and heat invading body of dog.The dampness and heat destroied the function of spleen.The complex prescription Kuqin was composed of scutellaria、kuhseng、chinese pulsatiila root and so on.According to theory of traditional Chinese medicine and clinic dialectical principle,to remove toxic heat and dampness through diuresis,treating dysentery and relieving hematochezia,arresting vomiting.It is the first time to study the function of traditional Chinese medicine on Canine parvovirus. We made a lot of tests of the complex prescription Kuqin,such as prevention and cure effect、immunity test、antibiosis test out of body、relieve fever、safe test and so on.First,According to prescription theory of traditional Chinese medicine and dosage form of clinical medication,we select out prescription which made from scutellaria、kuhseng、Chinese pulsatiolla root and so on.Then,we used the method which combined abstraction with water and precipitated with alcoh to extracted active component,and to prepared the medicine in injection and granules.As the same time,we gain the optimum condition by optimized the solubilizing agent, oxidation inhibiter,and filtering material,then research it’s quality.The analyze shows when adding 0.5%solubilizing agent（tween-80）,0.2%oxidation inhibitor（natrium bisulfurosum）,filter membrane and then filtrate,the injection appears marron and clear,no deposition,power of hydrogen between 6～8.,which contents of baicalin is 30mg/ml and matrine is 10mg/ml;the granules appears yellowish-brown,dry and uniformity,which contents of baicalin was 90mg/ml and matrine was 90mg/ml.Second,canine parvovirus model established,it was prevented and cured by the complex prescription Kuqin.Test showd that the granules had 90%protection rate and had 50%cure rate to dog.the injection has 90%protection rate and has 70%cure rate to dog.while Baiwu mistura only has 30%protection rate to dog.By observing the histological section of cardiac muscle,stomach and intestines,the complex prescription Kuqin can effectively prevent the dog frommyocardial damage, gastrointestinal hemorrhage,caused by canine parvovirus.The result proved that the compound Kuqin has a better preventive and therapeutic effect.Third,By test on cell counting,serum protein electrophoresis、macrophage phagocytosis test, determine the population of immunocell andγ-globulin,englobement percentage of macrophage、change of index of phagocytosis,The test shows that the complex prescription Kuqin can efficiently increase the quantity of WBC and achroacyte、histoleucocyte,also increase the content ofγ-globulin,enhance the englobement capacity for macrophag.By effecting Canine parvovirus,F81 bacteria has cytopathic changes,by observing the restrain function for bacteria cytopathic changes.Results shows the complex prescription Kuqin has a better restrain capacity for canine parvovirus.The IC₅₀is 1:587.5,Different times of dilution has better function on restrain the virus propagate,20 times of concentration of the has the maximum restrain on virus,Maximally tolerated dose test result showed,LD₅₀is 16.7g/kg,which is more than 100 times of clinical medication more safe and reliable.By making the bacteriostasis test out of body,the complex prescription Kuqin has a certain degrees of restrain function for E.coli、Staphylococcus aureus（SA）and so on.Test shows the result of E.coli was 3.75mg/ml、SA was 1.875mg/ml、Clostridieum welchii was 15 mg/ml、Salmonella and Blastomyces albicans 7.5mg/ml.By establish a rabbit fever model using,this medicine to cure heat release,test shows that in 2 hours later（high dosage）,the body temp can low down by 0.5℃,and it continued,after another 2 hours,the temperature comes to normal.Which has the similar function with injection bupleuri,another test on（low dosage）showed that the body temp can low down by 0.2℃in 2 hours later,and then come to normal in another 5 hours.The research showed that the complex prescription Kuqin has a better preventive and therapeutic effect on canine parvovirus.The complex prescription Kuqin can protect the heart, stomach an intestines,prevent from blooding,It also protect F81-cell from canine parvovirus.Enhance the quantity of achroacyte and histoleucocyte and restrain the pathological changes indust by canine parvovirus,It is ease the fever efficiently,and restrain the growth of E.coli、SA and other bacteria.It is safe and reliable in clinic.
