The Effects Of Environmental Factors On Growth And Growth-related Genes’ Expressions Of Amphiprion Ocellaris

Abstract/Summary:
We have studied the effects of different temperatures, salinities, photoperiods and color lights on growth and expressions of growth hormone receptor I(GHR1), growth hormone receptor II(GHR2),insulin-like growth factor(IGF)and insulin-like growth factor binding protein(IGFBP) of young Amphiprion ocellaris’ s liver to find out the optimum temperature and photoperiod of them. The results showed that there is no significant effect on the survive rate for young A.ocellaris at each temperature, salinity and photoperiod level, which are higher than 95% in 25 d and 90 % in 50 d.But in color light text, red light has the highest survival rate, which is 95±2.22% in 25 d and 92±1.92% in 50 d. However, blue and green light had the lower survival rate, which are 65.00±1.32%(25d), 35.00±1.92%(50d) and 53.30±2.33%(25d) 、36.70±3.22%(50d). On growth aspects, different temperatures, salinity, photoperiods and light color have greater impact on their growth. The results showed that young A.ocellaris had the highest growth at the 27℃group, higher growth at 22℃group and lower growth at 17℃group and 32℃group. In the salinity text, young A.ocellaris had the highest growth at 25 and 30 group, higher group at 20 group and lower growth at 15 and 35 group. In the photoperiod text, the young A.ocellaris had the highest growth at 24 h photoperiod group, higher growth at 16 h group and lower growth at 8h group and 0h group. In the light color text, red light had the great effect on growth, however, blue light and green light would reduce the growth rate. The molecular biology study began at the 50 th day, the expressions of GHR1, GHR2, IGF, IGFBP positively correlated with the growth. The result showed that the highest level of the three genes’ expressions were at temperature 27℃group and higher at 22℃group, the lower levels were at temperature17℃group and 32℃group. In the salinity text, higher growth at 20, 25, 30 group, lower growth at 35 group, lowest at 15 group. In the photoperiod text, that the highest level of the three genes’ expressions were at 24 h photoperiod group and 16 h photoperiod group. The study showed that temperature, salinity and light can effects the growth by controlling the GH/IGF growth axis, meanwhile, the results indicate that the most suitable temperature for young false clownfish are at 22℃-27℃,salinity at 25-30,photoperiod at 16h-24 h and red color is better for their growth.
