Bovine tuberculosis is a caused by Mycobacterium bovis in the respiratory system infection primarily zoonotic chronic consumption of infectious diseases. Has a broad host due to Mycobacterium bovis, cattle are the most sensitive host, people through without disinfection milk disease can be infected with TB, and tuberculosis and transmitted to cattle. Therefore, the cross infection between humans and animals is an important reason causing TB are widely spread. This is a very serious public safety problem which is urgent to be solved at home and abroad.At this stage, at home and abroad for bovine tuberculosis prevention and control of the main measures is “Quarantine-slaughter”, so timely and accurate diagnosis of bovine tuberculosis is the top priority of the prevention and control of bovine tuberculosis. OIE recommend for detection of bovine tuberculosis is the tuberculin test, this method is a based on the basis of type IV allergic reaction, with PPD and ot for anti original induced delayed cell hypersensitive reaction of intradermal test method. By mycobacterial cultures inactivated purified is PPD-B. Production process to Mycobacterium bovis strain, exists for the danger of poison, and with PPD-A, in the environment of widespread Mycobacterium and BCG have common antigens. Again, because of PPD is a mixture of a variety of protein, lipid and carbohydrate, qualitative and quantitative difficulty, batch to batch stability is difficult to maintain, test individual immune ability will affect PPD response level. Of course, in addition to the tuberculin skin test outside and such as antibody detection method, IFN-γ release method to diagnosis of bovine tuberculosis, but these methods is one of the key is to choose the most sensitive and specific reaction against the original auxiliary diagnosis. Therefore, with PPD as diagnostic antigen and its poor specificity, prone to false positive diagnosis and Mycobacterium bovis infection in the body in the process of secretion of a variety of antigens, using single antigen detection sensitivity is low, and recombinant fusion of protein components of clear, easy quality control, high specificity and biological safety risks and other advantages. So, in order to improve the specificity of detection and elimination of interference part of mycobacterial infection and BCG after inoculation, it is necessary to develop specific recombinant gene strong antigen.In view of the above reasons, this research adopts the experimental method of intradermal allergy based, We used BCG, Mycobacterium neoaurum, mycobacteria, Mycobacterium paratuberculosis and other different mycobacteria causing various immune guinea pigs,and then used the purified recombinant fusion protein MPB70-64, MPB83-70-63-64, MPB83-70-ESAT6-CFP10, ESAT6-CFP10, MPB51-63 and MPB63-64 and the PPD of 54 guinea pigs induced by DTH reaction, we will make the difference comparison between numerical measurements, there are four kinds of high specificity protein: MPB70-64, MPB83-70-63-64, MPB83-70-ESAT6-CFP10, ESAT6-CFP10, and MPB83-70-63-64 compared with the other three recombinant antigen has higher specificity, so the application of MPB83-70-63-64 as a reaction to the original 212 cows were detected by intradermal allergic reaction, and compared with the PPD, according to MPB83-70-63-64 as intradermal allergic reaction was used to detect antigen and detection of TB positive cattle and suspected positive cattle head number were lower than that of PPD as the number of head were detected, indicating that the antigen to improve the specificity of the detection of bovine tuberculosis. Therefore, MPB83-70-63-64 is expected to be used as an antigen for the detection of bovine tuberculosis, and play a role in the prevention and control of bovine tuberculosis.
