Artemisia argyi is the perennial herb of the genus Artemisia in the family of Compositae.It is a kind of natural herbal medicine which combines medical value and food nutritive value together.Moreover, it has a close connection with Dragon Boat Festival which is a traditional festival in China.It is a long-established tradition for Chinese people to collect Artemisia argyi and hang it on doors on Dragon Boat Festival. Because of excessive picking in the recent years,the wild resources of Artemisia argyi has been reduced rapidly,even in danger of extinction.In order to protect wild resources,the study focuses on the tissue culture and fast propagations system establishment of Artemisia argyi.Systematic study involves different factors which affect the process of tissue culture of Artemisia argyi.The study aims at indentifying the optimum media for the explant sterilization method, ideal medium for growth differentiation, and best way to root and transplant engraftment. The study helps to lay the technological foundation to protect wild resources, to meet the needs of seedlings and to establish the gene pool of Artemisia argyi.Major contents and results of the study are as follows:1. Explants disinfection sterilizationThe research results show that concentrations of HgCl2 and sterilization time affect foreign implant infection rate and survival rate. 0.1% HgCl2 leads to best sterilization result. The extension of soaking time leads to reduced infection rate and increased death rate. In the treatment of 6 to 8 minutes, the survival rate reaches the best.2. The cultivation of growing point differentiationThe results demonstrates that ideal growing point differentiation culture medium is MS + 6- BA 0.5 mg?L- 1 + sugar 30 g?L- 1 + AGAR 8 g?L – 1. Three division element of different concentrations can promote bud growth. 6 – BA leads to the best growth result. Plus, the growth rate is associated with the concentration of 6 – BA. 6- BA concentration of 0.5 mg?L – 1can better promote growth, with average height of 3.3 cm and more lateral bud. On the other hand, high concentration has a certain inhibitory effect.3. The cultivation of rooting and subcultureThe results show that MS + IAA 0.5 mg?L- 1 + sugar 30 g?L- 1 + AGAR 8 g?L – 1 is the ideal culture medium, and helps to achieve better root growth and higher germination. On the other hand, higher concentration of IAA(more than 0.75 mg?L-1)inhibits root growth. Because of the short and thick roots, less lateral roots grow.4. Transplanting and engraftmentThe results show that mugwort tube seedlings enjoy higher transplanting survival rate, nearly 90% and keep all the biological characters of wild plants. Three factors are taken into consideration, different types of cultivation matrix, the time of bottle refining, and usage of rooting powder. The results show that the mixed soil cultivation matrix, 6 days of bottle refining, and not adding rooting powder, lead to the highest germination rate.
