| The single nucleotide polymorphisms (SNPs) of MHC.SLC11A1 and BLF in 2296 Holstein cows of 22 dairy field in Ningxia province were tested by polymerase chain reaction, single strand chain polymorphisms (PCR-SSCP),and SNPs of EEF1D.TRAPPC9JHSP70.1 and TLR4 were determined by Sequenom. Analysis polymorphisms of gene variation the correlation between the different genotypes and somatic cell scores (SCS) in milk by least-squares means by molecular biology software,and the results are as follows:1.12 SNPs were detected in Seven candidate mastitis-related-genes:the mutation of C459T were observed in exon2 of MHC-DRA gene, one mutation of C7401G in exon 11 of SLC11A1 gene were found,and three SNPs (T916C,G926A,G928A) were observed in intron8 of BLF gene.The mutations of G1A and G3A were found in 5’UTR of EEF1D gene, a mutation of G2525852T in intron6 of TRAPPC9 gene were found, the mutation of G2409A were observed in coding region of HSP70.1 gene;rs29017188 in 5’UTR,rs8193046 in intronl and rs8193060 in exon3 were all found polymorphism of TLR4 gene.2.Association analysis of mastitis related genes and SCS showed that least squares mean of SCS for CC genotype was extremely significantly lower than that for GC genotype(P<0.01) in SLC11A1 gene,and the CC is mastitis resistance genotype; least squares mean of SCS for GA was extremely significantly lower than that for GG(P<0.01) in EEF1D-1 gene,and the GA is mastitis resistance genotype; least squares mean of SCS for GG was significantly lower than that for AA(P<0.05) in EEF1 D-3gene,and the GG is mastitis resistance genotype;least squares mean of SCS for GG was extremely significantly lower than that for TT(P<0.01),significantly lower than that for GT(P<0.05) in TRAPPC9-2525852,and the GG is mastitis resistance genotype.These results will provide an important theoretical basis for anti-mastitis individual breeding in Holstein Cow. |
