| The aim of this paper is to characterize the relevance between production traits and the variants of bovine hsp70 gene by the PCR-SSCP analysis of hsp70 5'-flanking region, to search the putative transcription factor binding sites in the 5' flanking region using the bioinformation methods, and to predict the feasibility of these variants .1 The PCR-SSCP analysis in 5'-flanking region of hsp70 geneFour fragments within 5' flanking region of hsp70 gene were amplified using the sequences registered in the NCBI to design the four pairs of primer. And using the PCR-SSCP, the nucleotide diversity and sequenced these variants were identified. The putative transcription factor binding sites within 5' flanking region were searched by the bioinformation methods. The results indicated that there be four fragments within 5'-region: P1, P2, P3, P 4 from upstream to downstream. We found that two, P2 and P4, had four base variations, 602 site T was lost, 691 site A->C, 1220 site A->G, 1309 site A->G Many putative transcription factor binding sites within 5' flanking region were found by using the Matlnspector software, including the ubiquitous transcription factors (SP1/GC elements and TATA box) and HSE(heat shock factor receptor ) . The mutation of 691site was in the region of HSE, and it may change the efficiency of transcription.2 The relevance between production traits and the variants of Holstein hsp70 geneBy the analysis of production traits, we found that there was no significant positive effect on production traits between AB genotype and BB genotype in P2. We also found that there was no significant positive effect on production traits between AB genotype,AA genotype and BB genotype in P4. But the temperature of rectum of AB genotype was lower than BB genotype in P2,though the difference was not significant. We predicted that A gene was more tolerant to heat stress than B gene. |
