Tan sheep fur is popular with people all over the world, but the quality of the wool determines the economic value of fur. For improvement of wool traits have been one of the important traits for Tan sheep breeding.This experiment selected the 296 Tan sheep in Ningxia as the research object, and chose two groups of genes were related to the traits of the growth of wool, which are MTNRlb、FGF-5 gene. According to the GenBank released these two gene sequences of sheep as a templatedesign 2 primer of strong specificity and high product yield for PCR-SSCP, and through the SAS software analysis of the two groups of gene polymorphisms and correlation of wool growth traits at the Tan sheep.The results of the study are as follows:There are two MTNR1b genotypes AA and AB were found by PCR-SSCP, PIC of MTNR1b value is 0.444between 0.25 to 0.5, belong to moderate level of polymorphism, which shows that MTNR1b gene has abundant genetic polymorphisms.In the two genotypes, the frequency of AA genotype was 0.336, AB genotype frequency was 0.664,accounts for about two-thirds of the population;A and B allele frequency of 0.668 and 0.332 respectively; After sequencing, the B allele of MTNR1b gene was found 244bp a base G missing.The AB genotype of MTNRlb gene was significantly correlated with Ningxia tan sheep wool length and wool bending number.There are two FGF-5 exon 1 genotypes AA and AB were found by PCR-SSCP,PIC of MTNRlb value is 0.402between 0.25 to 0.5, belong to moderate level of polymorphism, which shows that FGF-5 gene exon 1 hasabundant genetic polymorphism of. In the two genotypes, the frequency of AA genotype was0.444, AB genotype frequency was 0.556; A and B allele frequency of 0.722 and 0.278 respectively; The B allele of FGF-5 exon 1 gene was found the base C→A transversion at 52bp after sequencing,.The AB genotype of FGF-5 exon 1 geno significantly correlated with Ningxia tan sheep wool length and wool bending number.There are three FGF-5 exon 3 genotypes AA, BB and AB were found by PCR-SSCP,PIC of FGF-5 exon 3 value is 0.423between 0.25 to 0.5, belong to moderate level of polymorphism, which shows that abundant genetic polymorphism of FGF-5 gene exon 3.In the three genotypes, the frequency of AA genotype was 0.578, AB genotype frequency was 0.314,BB genotype frequency was 0.108, allele A frequency was 0.735. After sequencing, genotype of FGF-5 gene exon 3 was found 179 bp a transformation of a base A to G by sequencing. This SNP makes Gin into Arg.The AB genotype of FGF-5 exon 3 gene was significantly correlated with Ningxia tan sheep wool length and wool bending number.
