In this study, used 754 fine wool sheep individuals as research materials, screening the SNPs of FZD3、SFRP4 、 Notch1 and PTPN13 candidate genes by using combination of DNA pool with the second-generation re-sequencing technology; at the same time, screened out the SNPs which are may be significantly associated with wool traits; the two significant SNPs and nine SNPs of candidate genes were conducted typing by using Mass Array technology; Used SAS 9.1 least squares methods to analysis the correlation of 11 SNPs with wool traits, for the hope that screening of molecular markers which are associated with economic traits of fine-wool sheep, and also provide experimental evidence for the cultivation of new breed of fine-wool sheep.The main results were as follows:1. Testing the SNPs which are associated with wool traits of fine-wool sheep by using DNA pool technologyScreened out the SNPs which are associated with wool traits in the both very fine and very thick DNA pool by utilizing re-sequencing technology: T19381438 G and G19381838 T, distance between these two points in the 120 kb place of ACAN gene; FZD3 gene screened 32 non-synonymous mutation points, four synonymous mutation points and a password terminator; Notch1 gene screened three non-synonymous mutation points and nine synonymous mutation points; PTPN13 gene screened nine non-synonymous and nineteen synonymous mutation points; SFRP4 gene only found one synonymous mutation point in the coding region.2. Linkage disequilibrium analysis and haplotype analysis The two mutation points T19381438 G and G19381838 T of ACAN gene were in the strong linkage disequilibrium state, D’ and r2 values were: 1 and 1; the T101275261 C and A101276228 C, G101300776 T and T101275261 C mutation point of PTPN13 gene were in linkage disequilibrium state, D’ and r2 values were: 1 and 0.326, 1 and 0.469.3. Polymorphism analysis of candidate genes and its association with wool traits of fine-wool sheepAnalyzed the GLM of wool traits with 11 SNPs by using SAS 9.1 software, the results showed that the A101762198 T mutation point of FZD3 gene have a significant correlated with greasy Weight(P<0.05), A2886871 G mutation point of Notch1 gene have a significant correlation( P<0.05)and very significant correlation(P<0.01)with identification of body weight as well as weight after shearing respectively, A2899358 G mutation point have a significant correlation(P<0.05) with the fiber dispersion, variance coefficient and identification of body weigt; A101281906 C and A101279366 G mutation points of PTPN13 gene have a significant correlation(P<0.05) and a very significant correlation(P<0.01) with weight after shearing respectively. A101276228 C mutation point have a significant correlation(P < 0.05) with identification of body weight.
