The protein encoded by GAMyb was an activated transcription factor in gibberellin signalling, which plays an important role in promoting seed development. In this study, the specific primers were designed according the TaGAMyb sequence which located in 3A^ 3B and 3D chromosome and allelic variation and transcription express level were identified in order to invesgate whether the GAMyb is one of reasons leading wheat seeds with different depth of dormancy. Any allelic variations in TaGAMyb-A and TaGAMyb-D were not found. Two allelic variations were designed as TaGAMyb-Ba and TaGAMyb-Bb in TaGAMyb-B and were spread widely.Comparision to sequence of TaGAMyb-Ba, the 84 bp insertion was found in TaGAMyb-Bb by using the primer GAMyb-B2. A total of two population were used to validate the relationship between the polymorphic fragments of TaGAMyb-B and dormancy level. Statistical analysis indicated that GI values of two allelic variations of TaGAMyb-Ba and TaGAMyb-Bb were not associated with dormancy level in these two population (P value was 0.275 and 0.099, respectively, in Popu 1 and Popu 2), but GI values of two allelic variations of TaGAMyb-Ba and TaGAMyb-Bb were associated with different height in these two population in Popu 3, (P value was 0.000).To further determined the transcript level of TaGAMyb-A, TaGAMyb-B and TaGAMyb-D in cultivars with different depth of dormancy level and different height, Four cultivars Jimai 19, Xiaobaiyuhua, Zimai 12 and Neixiang 19 with different depth of dormancy level and different height were selected. Semi-quantitative RT-PCR analysis showed that:in the 30 day after flowering germination, second internode and leaf, the transcript expression level of Jimai 19 and Zimai 12 were expressed more highly than Xiaobaiyuhua andNeixiang 19, respectively, in TaGAMyb-A. in the 30 day after flowering germination, second internode and leaf, the transcript expression level of Jimai 19 and Zimai 12 were expressed more highly than Xiaobaiyuhua and Neixiang 19, respectively, in TaGAMyb-B. The transcript expression levels of TaGAMyb-D in four cultivars were expressed lowly, and expression level was minor difference in cultivars with different depth dormancy level and different height.
