Bacterial fruit blotch caused by Acidovorax citrulli presents a serious threat to watermelon, melon and other cucurbits industry. As a gram negative bacteria,A.citrulli harbors many RND multidrug efflux pumps protein coding genes.RND multidrug efflux pumps play an important role in the bacteria multidrug resistance and bacteria/plant interactions. In order to investigate the contribution of RND multidrug efflux pumps to antibiotic resistance and environmental adaption of A.citrulli,we studied their functions by constructing mutants and complementary strains. The results showed that RND multidrug efflux pumps related genes (marC、nodT、hlyD、rpsL) affected A.citrulli in the bacterial biofilm formation ability, ability to adapt to adversity, drug resistance, extracellular polysaccharides and extracellular cellulase secretion, growth ability.Relative expression of genes revealed that the copG gene expression was not affected by RND multidrug efflux pumps. And the expression levels of other related genes (hrcN、emrB、araC、ompR、 osmC、fliC were changed. This change may help bacteria restoring health.
