Melon production has been playing an increasingly important role in rural development and has become a pillar industry in China’s agricultural development.Bacterial fruit blotch of melon affects many melon crops in each stage,is a serious disease in melon production,has a great threat to the melon and fruit economy of the global bacterial disease,mainly harm watermelon,melon leaves and fruit,produce a large number of disease spots and fruit decay,often causing devastating consequences,is one of the most important crop diseases in recent years.Nevertheless,research on BFB has been limited to date.This study successfully build the Acidovorax citrulli fluorescent strains mcherryXJL-12,the strain can stably express red fluorescence,has been used to internal in Acidovorax citrulli into the seeds of inquiry.This study used marker strain mcherry-XJL-12 in the position of colonization and colonization of watermelon in the body,Acidovorax citrulli infect the host place,process,and will be the study of colonization in the watermelon.The experimental results show that the mark strains in watermelon root surface adsorption and enrichment of first by the root epidermis microporous or wound,and then into the root endodermis cells and vascular tissue,through the vascular bundle,along with the water and minerals up to stems and leaves,at the same time,mark strains can also colonization in watermelon plants,such as leaf epidermal cells,endothelial cells,vascular bundle,intercellular space,porosity in areas such as the surrounding tissues.Therefore,this paper makes a preliminary study on the way of melon fruit spot pathogen entering the seed through the root,which has important theoretical significance and potential application value for the research on the infection route of melon bacterial fruit spot pathogen,and has important guiding significance for the prevention and treatment of this disease.Bacterial fruit blotch(BFB)is a seed-borne disease.With a long history of watermelon cultivation,China is the world’s largest watermelon and melon producing country,and also the seed production base of many multinational seed companies.In recent years,Acidovorax citrulli fruit spot disease has occurred in many provinces of China,bringing great losses to local farmers and seed producers.Therefore,in this study,94 samples of cucurbit rootstock seeds(pumpkin,zucchini,pumpkin rootstock and zucchini rootstock),762 samples of watermelon seeds and 308 samples of melon seeds from different regions in China were monitored for Acidovorax citrulli carrier status in 2017-2018 using by PCR and Bio-PCR technique based on pathogen isolation.From 2017 to 2018,55 and 39 seeds of gourd rootstocks were collected from all over the country in this study,and the germborne rate showed a decreasing trend,which was 9.09%and 5.13%,respectively.Among them,the highest detection rate of Acidovorax citrulli bacteria was 16.67%in Xinjiang origin.In addition to cucurbit rootstock seeds,watermelon and melon seeds were also tested in this study,and the detection rates of watermelon acid bacteria were 5.77%and 11.36%,respectively.The experimental results proved that Acidovorax citrulli were spread from pumpkin rootstock seeds to watermelon seedlings by grafting,thus causing watermelon seedlings to wither,indicating that infected gourd rootstock seeds may be an important source of infection for Acidovorax citrulli. |
