Purpose:The fatty liver desease of Tilapia happen frequently in the feeding phase,that immensely hinder the development of breeding industry of Tilapia. The experiment establishs the fatty liver desease model of Tilapia by increasing the feeding level,filtrate and identify the differently expressed proteins between the healthy Tilapia and fatty liver desease model by the iTRAQ-LC-MS/MS technology,deeply analyze the effect of differently expressed proteins to the lipid metabolism in the process of the fatty degeneration,establish new thinking for detection and prevention of fatty liver desease.Method:According to preliminary study,design the feeding level of experimental group B for 10%,control group A for 6%,then verify the fatty liver desease model by testing the HSI,lkiver lipid content,related biochemical criterion and liver tissue slice after feeding eight weeks.Immediately choose 36 fishes from every group,mix 36 liver sample half-and-half,mark the sample by iTRAQ after protease-restricted;separate the liver proteins by LC-MS/MS technology,and use ProteinPilotTM Software 5.0(AB Sciex)for retrieving proteins data,then get the credible and quantifiable proteins,based on the false discovery rate(FDR)<0.01, setting ratio between two groups Ratio(B/A)>1.3 or Ratio(B/A)<0.77 for screening up/down differently expressed proteins,then analyze the differently expressed proteins by GO classification annotations and KEGG Pathway,and get the fatty liver related objective proteins by combining above result with references.Results:Got 166 differently expressed proteins by setting Ratio(B/A)>1.3 or Ratio(B/A)<0.77,thereinto 78 is up expressed proteins,down expressed proteins found 88;these proteins participate in 37 biological processes cantain transport,small molecule metabolic process,carbohydrate metabolic process,lipid metabolic process,biosynthetic process,catabolic process,response to stress, lipid binding and else,thereinto 29 proteins are closely related to fatty liver desease,and the up expressed proteins contain RAC1,ATP-citrate synthase,Acetyltransferase component of pyruvate dehydrogenase comple-x,L-F ABP,ADRP,SOD,ACSL,Trx,ICDH,GP,GLUT2,GST and HADHA;down expres-sed proteins contain COX4,HMG-CoA,HL,CYP7A1,APOA4a,AOPA4b,AOPB-100,LCP,CLU,CYP51,SPARC protein, VTQcomplement factor B, complement C5, FPPS and ACBB11.Conclusion:By the means of GO classification annotations and KEGG Pathway analyzing the 29 objective proteins,the result show that the expressed level of these objective proteins promote the liver lipid synthesis,inhibit the stea-tolysis,obstruct the transport of lipid,trigger the inflammatory response of liver t-Issue and induce the insulin resistance,respectively.Thus expedite the formation of fatty liver model.The research reveal the pathogenesis of fatty liver desease f-rom proteins level,provide a new idea for curing the fatty liver desease.
