| Butterfly orchid(Phalaenopsis amabilis) is a kind of perennial herbaceous attached-living orchid in the tropics. It was named butterfly orchid for its butterfly-like pattern. As a kind of popular upscale flower in market at home and abroad, it is deeply loved by people around the world and has extremely high ornamental value and economic value. The vegetative and seed propagation of butterfly orchid are difficult to meet the needs of mass propagation. And its aerial roots have high requirements for cultivation matrix’s permeability and water-retaining property. For the demands of the market to good varieties and new varieties, the tissue cultivation and plant technique of butterfly orchid have become important research topic. This paper mainly studies the plant effect of induction culture, multiplication medium and matching different cultivation matrix to explore culture medium of tissue culture in butterfly orchid, improve the efficiency of propagation, and to look for a kind of substrate with good effect of cultivation which can take the place of high-cost water moss. We can draw the conclusions through the study that:1. Choose young peduncles of blossomed butterfly orchid as explant carrying on the induction culture. The explant can achieve the good effect of lower rate of pollution when it is processed with 0.1% Hgcl2 for 12 minutes; The suitable concentration of 6-BA for induction culture of axillary buds is 9 mg/L(1/2MS as basic medium, NAA 0.3 mg/L). The appropriate culture medium of multiplication culture is 1/2MS+6-BA9mg/L+NAA0.3mg/L+ CM100ml/L with which the effect is the best, and buds are robust.2. Organic wastes such as bark, sawdust, etc. can substitute water moss to be the cultivation matrix of butterfly orchid. According to the analysis of the growth and morphological index of butterfly orchid, the effect of the group(tree bark: corn cobs: sawdust5:3:2) is the best than other groups, which can replace water moss to be the cultivation matrix of butterfly orchid. The effect of the group bark: water moss(1:1) is secondly good.3. The result of cost accounting shows that the use of organic wastes as substrate material can drastically reduce the cost of cultivation. Under the premise of normal growth of butterfly orchid, the mixed matrix with bark, water moss and sawdust can be used as the cultivation matrix of butterfly orchid to greatly reduce the production cost. |
