In this research a lesion mimic wheat mutant LF2010 which was obtained from an anther culture line H261 that mutated with EMS was chosen as material, and different gene mapping strategy was taken to map the new lesion mimic gene lm3. The main results were as follows:1. Genetic analysis: A cross was made between the mutant LF2010 with the normal wheat line Chinese Spring. Genetic analysis was conducted with F1, F2 population and F3 lines respectively. The results showed that the spots traits was controlled by a recessive gene in the mapping population of LF2010 and CS.2. Gene mapping: When BSA gene mapping strategy was taken, only a marker Xwmc388 that linkage with the target gene was obtained. Because of Xwmc388 had multiple sites on wheat chromosome, chromosome of gene lm3 could not be determine initially; Further MBSA gene mapping strategy was utilized, 2 marker Xwmc417, Xbarc134 that linkage with the gene lm3 was obtained. Finally the target gene lm3 was mapped on 6B chromosome.At the same time, strategy combine wheat 90 K microarray gene mapping and BSA was used, the number of SNP that was distributed on each chromosome was counted between F2 two trait pools based on the integration of high-density 90 K SNP genetic linkage map. This approach could be map gene lm3 quickly on wheat 6B chromosome. According to the results of the enriched interval of wheat 90 K chip, SSR primers based on wheat genome were designed; and 3 SSR primers that linkage with the gene lm3 was obtained. It was further validated that the target gene is located on chromosome 6B.After population verification, it was obtained that a genetic linkage map that included 15 pairs marker linked with lm3. Finally lm3 was located on 6BL of wheat chromosome, 13.8 cM away from marker SWES2 and 3.8 cM away fromXbarc134.
