In the male mammal mature individuals, the formation of sperm for the continuation of the species is a very important mechanism for self-renewal. It bears the role of the formation of male gametes and gene transmission which is necessary for evolution. Now puberty, the spermiogenesis throughout the whole life course, produce large amounts of sperm by the orderly and strict regulation of cell proliferation and differentiation progress, which is taken place on the basis of spermatogonial stem cells (SSCs) self-renewal. As male germ stem cells, SSCs are A-single cells produced on the spermatogenic epithelial basement membrane of stesticular seminiferous tubules, whose precursors are undifferentiated stem cells. Spermatogonial stem cells can self-proliferation aspect update itself remained relatively constant number, on the other hand, SSCs can be subjected to several postmitotic enter meiosis, the formation of primary spermatocytes eventually form sperm. SSCs can remain number in a relatively constant state in the way of self-proliferation, on the other hand, after numbers of mitosis it enters meiosis, companying the form the primary spermatocytes and sperm eventually.Glial cell line-derived neurotrophic factor (GDNF) is produced by capra hircus epithelium cells of the seminiferous tubules, which play a decisive effect on the proliferation and differentiation of self-renewing SSCs, through the collaborating with glial cell line-derived neurotrophic factor receptor alpha 1 (GFRa-1). we have not did the deep research about the gdnf gene of the capra hircus prepubertal testis nor its receptor gfra-1. Therefore, this experiment is devoted to the study of capra hircus testis gdnf gene and the expression of its receptor gfra-1. As well as their effect on the variation of SSCs and changes of the structure testis in the process of development.We chose 11 groups of materials which are the same part of testis tissues from pre-pubertal capra hircus bear after 45、51、56、66、69、73、86、95、102、110、122 days respectively.Firstly, for the part of gene, we used the application of RT-PCR and real-time PCR methods to analyze the amount of two genes transcription of mRNA of different birth age capra hircus testis. Secondly, we used the application of SDS-PAGE、 Western blotting and the analysis of protein structure of bio informatics to analyze the expression amount of GDNF and its receptor GFRa-1 in different age born capra hircus. Finaly, we use the application of HE staining to stain testis structure, for pictures of capra hircus testis structure. We have three results:(1) The expression of gdnf gene and gfra-1 gene shows the same change trends in the puberty capra hircus testis for the same born date. The expression of the two genes is reached the peak for capra hircus testis born 86 days. For only one gene, both genes expression of capra hircus testis tissues are Reciprocating change.(2) The expression of GDNF and GFRa-1 shows the different change trends in the puberty capra hircus testis for the different born date. The expression of the two genes is reached the Minimum value for capra hircus testis born 86 days. The expression of GDNF is down at 102 days, while GFRa-1 is up.(3) Pictures show that the greater the day age, the thicker the lumen of the tubules in the testis structure, the less the epithelial cells. Part of the stem cells differentiation, suggesting the capra hircus testis development is gradually mature.
