Ulcerative colitis is a kind of harmful inflammatory bowel disease, the researchers found that Lactobacillus acidophilus can play a role in the treatment of this disease by TLR2 signaling pathways. Inflammatory bowel disease has become an important disease in veterinary clinical. So research the effect and mechanism of Ch TLR2 signaling pathway in ulcerative colitis chicks prevented by Lactobacillus Acidophilus. The colitis model chicks were used in this study, using detection methods such as HE, immunohistochemistry, ELISA and Real Time PCR to detect the expression levels of TLR2, MyD88, IL-8 m RNA and TLR2 protein content in colon tissue, IL-8 and IL-10 protein content in serum, the clinical signs and pathological changes of Ulcerative colitis modeling chicks(T group), Lactobacillus acidophilus pretreatment modeling chicks(L group) and the control chicks(C group). The results showed that:(1) Enema 15-day-old chicks by TNBS and ethanol compound, can successfully replicated in animal models of ulcerative colitis.(2) pathological lesions of chicks mainly in the caecum, appendix, colon and cloaca, and the level of inflammation of chicks in group T are higher than chicks in group L.( 3) It is 1 to 3 days and 1 to 7 days after chicks was given TNBS ethanol mixture through the gut of them, the expression levels of TLR2, MyD88, IL-8 mRNA in colon tissue of chicks in group L are various degrees lower than the chicks of group T, group L and T are higher than the control chicks.(4) after clyster, the expression levels of IL-8 mRNA in colon tissue of chicks in group L are various degrees lower than the chicks of group T, IL-10 protein expression levels in chicks serum of group L are various degrees higher than the chicks of group T.(5) Immunohistochemistry show that: TLR2 protein in colon tissue of UC chicks, was mainly distributed in the inflammatory mucosal, and with the colon pathological changes aggravated, TLR2 protein expression was also increased. The results show that the expression levels of TLR2 signaling pathways associated factor and inflammatory pathological damage in group L chicks are significantly lower than T grou p chicks. Lactobacillus acidophilus can reducing the release of IL-8 by inhibiting the over-activated of TLR2/MyD88 signaling pathway, reduce inflammation the damage to chicks.The experiment studied on the role of Ch TLR2 signaling pathway and related fact ors in UC chicks that Lactobacillus acidophilus prevented, provide important referenc for revealing Lactobacillus acidophilus start and regulat the mechanisms of immune response based PPRs signaling pathways, and an important theoretical basis for the prob iotics used in the prevention and treatment of related diseases.
