In this study, we used 15 day-old chicks as the research object, in the success of preparing ulcerative colitis chicks model based and applied immunology, traditional pathology inspection methods and modern molecular biology techniques, through to count the number of the total number of leukocytes and different types of leukocytes in chicks’ peripheral blood, and test the dynamic changes of immunoglobulin IgA, IgM and IgG and cytokine IL-6 in chicks’ serum. Research aims to comprehensively and systematically expound the change of immune cells, immunoglobulin and cytokine levels in suffering ulcerative colitis chicks peripheral blood, further reveal the occurrence and development of chicks ulcerative colitis, in order to provide scientific experimental basis for prevention and treatment of other animal’s UC, especially human UC. Our results were as follows:(1)The mixture of TNBS solution and dehydrated ethanol at the rate of 1:1 was used to give the enema to chicks through anus, this can successfully replicate the model of ulcerative colitis of chicks.(2)chicks with TNBS and alcohol mixture modeling, UC model chicks During 3 to 11 days the chicks body weight of model chicks were significantly or extremely significantly(P <0.05 or P <0.01) lower than the control chicks,During 1 to 3 days weight loss, During 5 to 11 days weight showed a slow growth.(3)chicks with TNBS and alcohol mixture modeling,UC model chicks. During 1 to 9 days the total number of leukocyte and neutrophils, lymphocytes, monocytes of model chicks peripheral blood were significantly or extremely significantly(P <0.05 or P <0.01) higher than the control chicks; during 1 to 7 days, compared with the control chicks the total number of macrophages basophils of model chicks were extremely significantly(P <0.01) higher than the control chicks. Overall, during 1 to 3 days after the modeling the number of above inflammatory cells increased rapidly, when reaching the peak and then gradually decreased, while, the number of T and B lymphocyte compared with the control chicks, during 1 to 9 days after the modeling, all were significantly(P <0.01) higher than control chicks.(4)During 1 to 9 days after chicks with TNBS and alcohol mixture modeling, the content of immunoglobulin IgA, IgM and IgG of model chicks peripheral blood all were significantly(P <0.01) higher than the control chicks. No significant difference compared with control chicks at 11 days, restoration of the content to normal levels of the chicks.(5)During 1 to 7 days after chicks with TNBS and alcohol mixture modeling, the content of cytokine IL-6 of UC model chicks peripheral blood were significantly(P <0.01) higher than the control chicks, then it returned to normal levels, and no significant difference with the control chicks during 9 to 11 days after the modeling.
