This paper study on spent Ganoderma luncidum JUNCAO substrate(SGJS), research the extracting process, the quality control of extract, antioxidant activity of polysaccharides,the components of polysaccharides and application of extracts in the dairy cows feed.The mian results are as follows:1.Through single factor experiment and orthogonal experiment to explore extraction process of SGJS, the result showed that the optimal process for SGJS extraction is:extract at 100℃ for 2 h, extract 1 times, solid-liquid ratio is 1:10, and soak it for 12 h; Through verification, the yield of extract is 22.89%, the yield of crude polysaccharides is 10.91%.2.Quality stability of extracts is a important factor for the application research.Therefore, through testing the content of polysaccharides, protein, water and crude ash in different batches extract, to preliminary refine the quality standard of extract. As the polysaccharide is the active ingredient in the extract, so refine the content of crude polysaccharides is≥39%, the content of polysaccharides is≥9%.3.Antioxidant activity is one of the major bioactive of polysaccharides, the results show that hydroxyl radical clearance rate and reducing capacity have concentration-response relationship to the concentraton of polysaccharides, but the clearance of superoxide radicals is not observed.4.Through graded alcohol precipitation, to get 20%,33.3%,50%,66.7%,20% grade polysaccharides, and determined the antioxidant activity of different fraction polysaccharides. As results,20% grade polysaccharides shows the stongest hydroxyl radicals scavenging capacity; 33.3% grade polysaccharides shows the the strongest reducing capacity; The clearance of superoxide radicals is not observed.5.Each grade polysaccharides was isolated with dextran gel G-100, the elution curves show that each grade polysaccharides only has one main peak, which can be learned that, each grade polysaccharides has been purified.6.The water extract from SGJS was applied as feed additives in dairy cows, adding amount of 170 g/head/d for 30 days, the result shows that all the indicators are no significant differences, however, has a certain role in promoting. SGJS extracts can improve the milk yield of cows to a certain extent, and maintains the dairy products quality steady, has a regulatory role to blood biochemical indicators; In the experimental group, the content of immunoglobulins in milk are trending up, shows that the extracts has the certain promote role to the immunoglobulins content in milk; Compared with the control group, Immunoglobulins levels in serum present a rising trend, the experimental group IgG levels relatively increased by 13.48%, IgM is relatively increased by 10%, IgA is relatively increased by 4.11%; According to the results of serum antioxidant activity, the experimental group has improved significantly, compared with the control group, T-AOC relatively increased by 23.61%, GSH-Px activity relatively increased by 3.94%, the content of MDA relatively reduced by 4.91%,and the activity of SOD had no obvious effect.
