Study On Morphology And Embryo Culture Of Mature Lotus

The lotus flower is ornamental, edible rhizome.Lotus seeds can be used as medicine.The whole body is treasure.It is loved by the people.In order to avoid the disadvantages of traditional mode of reproduction,people began to study the lotus plant tissue culture with the plant tissue culture technology development.In this research,explants was mature embryos.Using 3 kinds of disinfectant disinfectant were 0.1%HgCl2,10%NaClO and 10%H2O2.Disinfection time of ea c-h disinfectant sets for 8min,6min and 4min.The basic medium was MS, whic h was added to plant hormones (6-BA and NAA).The 2 kinds of plant hormon e-s’s ratio was 1.And it was set up a total of 4 concentration gradient.The test results show that the 0.1%HgCl2 for 8min was mor appropriate to disinfect th e mature embryo. The contamination rate was O.The survival rate was 85.99%. T-he germination rate was 91.86%.The optimum medium was MS+0.5mg·L-16-BA+0.5mg·L-1 NAA.Tissue culture seedlings’growth was normal.When the 0. 1%HgCl2 was used to disinfect the mature embryo for 8min,culture medium w-hich was MS+1.0mg·L-16-BA+1.0mg·L-1 NAA could induce more root num-ber,bud number, and differentiation yellow grain of mature embryo number,foll-owed by 9.67,8.67,6.05;The explant was the germ and differentiation of som-e yellow particles hypocotyl which was induced by the mature embryo for 21 d.The inoculation to the successive transfer culture was MS+0.5mg·L-16-BA +0.5mg·L-1 NAA.The result was enlargement or extension of the germ was th-e results of successive transfer culture was enlargement or extension of the ge-rm,dead of the differentiation of some yellow particles hypocotyl,browning rate of 0,the average pollution rate of 60% in the germ successive transfer culture, the average pollution rate of 0 in the differentiation of some yellow particles h-ypocotyl successive transfer culture.Differentiation of some yellow particles hy-pocotyl was inoculated to the culture medium which induced the shoot.The res-ult was all the explants were death,browning rate of 0, the contamination rate of 0;The paraffin sections’s material was hypocotyl yellow particles which was induced by the mature embryo for 21 d and the mature embryo.It was observe d by the optical microscope.Tests found mature embryo cells were stained red. Hypocotyl yellow particles were stained green.It may be the parenchyma cell.
