CRTC(CREB Regulated Transcription Coactivator) is a conserved eukaryotic protein family, including CRTC1, CRTC2 and CRTC3. CRTC regulate transcription factor activity of CREB. Studies have shown the CRTC can integrate multiple signal paths. CREB-CRTC pathway plays an important role in the the process of hormones and nutrition on the energy balance.Using the biological software and online biological, we analyzed the characteristics of CRTC families of DNA and protein of cattle. The 13 different tissues expression of CRTC1 、 CRTC2 、 CRTC3 were analyzed by the method of Real-time-PCR, while the polymorphism of CRTC1、CRTC2、CRTC3 genes in Qinchuan were analyzed by PCR-RFLP and DNA sequencing methods. The correlation analysis of growth and carcass traits were analyzed by SPSS. In order to supply the scientific proof for the selection of Qinchuan cattle to own the better growth and carcass traits, the CRTC genes were selected in our research.The main results are as followed:1.The real-time polymerase chain reaction(RT-PCR) results of CRTC1 gene in Qinchuan cattle showed that among all types of tissue that were analyzed, the highest mRNA expression levels of the gene were found in lung 、 subcutaneous fat 、 spleen and kidney;middling expression were found in liver、heart、abomasum、muscle and large intestine;low expression were found in small intestine、omasum、reticulum、rumen、abomasum and heart.2.The real-time polymerase chain reaction(RT-PCR) results of CRTC2 gene in Qinchuan cattle showed that among all types of tissue that were analyzed, the highest mRNA expression levels of the gene were found in lung、subcutaneous fat、spleen and liver;middling expression were found in large intestine、small intestine、reticulum、rumen、muscle、intestine、abomasum;low expression were found in heart.3.The real-time polymerase chain reaction(RT-PCR) results of CRTC3 gene in Qinchuan cattle showed that among all types of tissue that were analyzed, the highest mRNA expression levels of the gene were found in lung 、subcutaneous fat 、 kidney and spleen; middling expression were found in liver、muscle、 large intestine、 small intestine、reticulum、rumen、intestine and abomasum;low expression were found in heart.4.Sequencing in the 275 individuals of Qinchuan cattle, two single nucleotide polymorphisms(SNP1 g.68206 G>A and SNP2 g.68230C>A) in exon 8 of CRTC1 gene were identified. In addition, two loci and H3H3(GGAA) diplotypes were significantly associated with some of the growth traits in Qinchuan cattle,5.Sequencing in the 302 individuals Qinchuan cattle, three single nucleotide polymorphisms(SNP1g.3001C>T、SNP2g.3034G>A and SNP3 g.3467 T>C)in the CRTC2 gene were identified.The association analysis showed that the three loci and H2H4(CCAGTT)diplotypes were significantly correlated with some growth and carcass traits6.Via the mothod of PCR-RFLP in 395 individuals of Qinchuan beef cattle,four single nucleotide polymorphisms(SNP1 g.62652A>G、SNP2 g.62730C>T、SNP3 g.66478G>C and SNP4 g.91297C>T) in CRTC3 gene were identified. In addition, four loci and H1H1(AACCCCTT)diplotypes were significantly associated with some of the growth and carcass traits in Qinchuan cattle.
