Gastrodia elata is a perennial herb, its tuber is a well-known Chinese herbal medicine. Gastrodia elata is highly degraded, it has no roots and leaves. It can’t produce into organic matter by the way of photosynthesis. In the growing cycle of Gastrodia elata, it’s necessary to establish a symbiotic relationship between two fungi. To investigate the relationship between stem tuber mass and seed production of Gastrodia elata f. glauca and educe the most suitable Gastrodia elata f. glauca weight for seed production, 200 g, 100 g, 80 g, 50 g, 30 g level stem tuber of Gastrodia elata f. glauca were bought from Xiaocaoba town, Yiliang County, Zhaotong City, Yunnan Provence and planted in plastic basin in a room to produce sexual seed. Artificial cross pollination for each flower. Results show that: the greater stem tuber produce more flowers, fruits, and seed after bolting. Unit weight of 200 g level stem tuber with the least(2.265 g / 500 g) seed yield, the lowest(85%) bolting rate and performance for bolting difficult, often late bolting and the difference in individuals bolting time larger(time of 21 days). As prices are much higher than other level stem tuber, 200 g level stem tuber used for seed production is not economical and feeling wasteful. it is not recommended to use. 100 g class stem tuber bolting rate of 95%, and no obvious bolting difficult phenomenon and the unit weight of the seed yield(2.850 g / 500 g) was significantly higher than that of 200 g and 80 g(2.500 g / 500 g) class stem tuber. Fitting for seed production. 80 g class stem tuber bolting rate of 100%, bolting time convergent. It already does not belong to the ideal merchandise stem tuber but a bargain, so the selection of 80 g class stem tuber are more economical and in a certain extent is a shift from inferior to superior. 50 g class stem tuber bolting rate was 100% and bolting time convergent. Seed yield of unit weight stem tuber is highest(3.180 g / 500 g), but some fruits smaller and even some fruits are not full with less seed and the reproductive capacity of the seed uncertain. 50 g class stem tuber should be carefully selected. 30 g level stem tuber shows weak bolting ability, does not recommended for use. In practice, for the sake of convenience, stem tuber weighted from 80 g to 100 g is recommended for seed production. To investigate the relationship between the different mass of stem tuber that belongs to Gastrodia elata and the germination rate of the sexual seed that produced and investigate the germination of the sexual seeds of Gastrodia elata after maturing. We immediately measure the germination rate of the sexual seed. First Mycena osmundicola is activated on the MEA plate medium. Then Mycena osmundicola is inoculated to FGM medium, shaking FGM medium to expanding culture and latter inoculate the Mycena osmundicola to MC culture medium to culture. Mycena osmundicola infestate the square deciduous leaves of Cyclobalanopsis glauca(Thunb.) Oerst whose side length is 2 cm. Puting the contaminative leaves on the center of water agar whose diameter is 9 cm.And finally spreading 100 sexual seed around the leaves on the water agar. The results showed that there is no significant difference in the germination rate of sexual seeds that is produced by the Gastrodia elata stem tuber that has different masses. To investigate the relationship between germination rate of sexual seed Gastrodia elata and different storage time, using the above method and testing the germination of sexual seeds that have been kept from0-24 weeks, 28 weeks, 32 weeks, 36 weeks, 40 weeks, 44 weeks, 48 weeks, 52 weeks separately. The results showed that: germination rate is 29.08% at first, the germination rate of the sexual seed Gastrodia elata from 1 week to 5 week is between 21% and 24%; the germination rate of the sexual seed of Gastrodia elata is decreased from 6 week to 9 week and its germination rates are respectively 13.5%, 7.5%, 1.45%, 0;there is no germination from 9 weeks to 17 weeks but a few sexual seeds of Gastrodia elata can restore its ability of germination at the week of 18.and its germination rate is 1.5%. The germination rates of the sexual seeds from the week that is 19 to the week 22 are increasing and respectively are 5.17%, 13%, 19.75%, 21.75%; The germination rates of the sexual seeds of Gastrodia elata from22 weeks to 52 weeks is between 20% to 22%.
