Moso bamboo is the most widely distributed and largest area with the largest proportion bamboo species in Chinese forest resources. It plays an very significant role in the production of bamboo industry with extremely high economic, ecological and cultural value. Bamboo grow very rapidly. In the natural state, it only takes the unearthing young bamboo 45 to 60 days to grow 20 meters high. When it comes to its growth peak period, the growth height in a night is expected to reach more than 1 meter. The fast growth rate is extremely rare in other non-bamboo flora.Previous stduy has gained some bamboo fast growth related structures, hormone levels, specific genetic information by the use of microscopy, high-throughput sequencing and proteomics research approach. But bamboo genetic transformation system has not yet been established, advantage gene function of bamboo is need further explore and research systemly.In 2009, Dr Minami Matsui in the institute of physical and chemical of Japan, has built a rice full-length cDNA library and made it over express in arabidopsis with a large-scale, to research the rice gene function, and named it as FOX (Full-length Overexpress) expression system. In addition, our country important forest species such as pinus massoniana and cunninghamia lanceolata, grow slowly and has a long cycle. How to shorten the production cycle and improve growth speed has always been an important objective of forestry production in China. Therefore, screening and develop the common fast growth bamboo genes is of great significance.Therefore, this project use the latest In-fusion technology and key DSN enzyme (Duplex specific nuclease), researching and developing abamboo full-length nomalization cDNA library independently, and make it overexpress in dicotyledonous plants arabidopsis with a random and large scale through horizontal gene transfer technology, hope to screen the fast growth genes of single or double cotyledons plants.By the use of bamboo gene expression on arabidopsis strain,40 specific leaf form,10 specific stem,23 short and small,12 early flowering,25 delayed flowering,10 fast growth and other phenotypes have been screened.36 bamboo gene has been gained by the comparison with phenotypic correlation in arabidopsis, further screening the single copy of inserted homozygous mutant, hope to screen of single or double cotyledons rapid growth genes, discuss the molecular mechanism of plant growth, providing oretical basis and accumulating resource for subsequent applicationon on forestry growth.
