By exploring human interference on polymorphism of Acanthopanax giraldii Harms to filter new materials that meets the developing needs, and by exploring the pattern of cutting interference to the endogenous hormone to provide the basis for promoting the germination and cultivation, this experiment was carried with Acanthopanax giraldii Harms in Aba Sichuan province. For better protection, utilization and development, RAPD molecular markers and high-performance liquid chromatography (HPLC) analysis was used to analysis the samples. The main results were as follows:1. In the uncutted group, the content of ZT and GA in terminal bud were higher than that in the lateral bud on average. The content of IAA and ABA of the lateral bud were higher than that of the terminal bud in the initial stage and lower later. In the mild cutted group, ZT content were respectively 87.3% and 43.4% of the lateral bud in the growth period. The content of GA of the terminal bud was 136.0%,131.7% of that in the lateral bud. The content of IAA of terminal bud and lateral bud went down rapidly, then the content of IAA in secondary terminal bud accumulated and reached 9.80 and 1.73 times of that of the lateral bud. The content of ABA in the terminal and lateral bud both went down, but the content of ABA in the terminal bud went down a little bit slower, relatively. The content of ABA in the terminal bud was 81.6%、608.5%、216.2% of that in the lateral bud. Declaring that mild cutting could eliminate the apical dominance in Acanthopanax giraldii Harms and was coducive to the germination of the lateral bud.2. In the uncutted group, the GA/ABA and the ZT/IAA value of the terminal bud and the lateral bud were vicissitudinous. In the mild cutted group, the GA/ABA values in the terminal bud were 447.6% and 164.1% of that in the lateral bud. The ZT/IAA value in the terminal bud was respectively 1.08,2.32,1.16 times of that in the lateral bud in each period. Mild cut made the GA/ABA value of the lateral buds rice and IAA/ZT values decline. And the variation tendency was on the contrary.In the heavy cutted group, IAA/ZT values of the terminal bud were also higher than that of the lateral bud, which were respectively 2.09,1.86,1.31 times of that of the lateral bud. In the same way,heavy cutting also made ZT/IAA value of the terminal bud rise. The results showed that mild cut broked the prime hormone balance in Acanthopanax giraldii Harms and established a new balance, which was advantageous to the Acanthopanax giraldii Harms to the growth and reproduction of Acanthopanax giraldii Harms.3. According to the polymorphism analysis of Acanthopanax giraldii Harms Under the artificial interference,15 RAPD primers amplified out 155 bands, in which there were 137 had genetic polymorphism. The polymorphism rate were 88.39%, the average number of alleles was 1.8839, the average number of effective alleles was 1.5112, the expected heterozygosity was 0.3020,and the Shannon diversity index was 0.4548. The data displayed that there was rich diversity among the samples, which provided the basis for cultivar identification and genetic breeding in the future. By means of cluster analysis,20 copies of materials were divided into four categories, which was consistent with the phenotypic traits. The genetic similarity coefficient was between 0.53~0.84. The largest genetic similarity was between sample 13 and 15, between which the genetic similarity coefficient was 0.8387. The smallest genetic similarity was bettwen sample 6 and 10, between which the genetic similarity coefficient was 0.5290. Therefore there was rich diversity among the the selected materials.
