In this paper,Chestnut (Castanea mollissima) was viewed as the research object,endogenous hormone content variation of the chestnut buds in different periods wereconsidered using the method of a combination of laboratory analysis and field trials, andthe impact of different fertilization methods were studied to endogenous hormone contentsof chestnut. the main findings are as follows:1. SPE-HPLC-based Endogenous hormone detection methodsThe qualitative and quantitative analysis methods of the four endogenous hormonesIAA, ABA, GA3and Z in chestnut were established in the test. The results show that,80%precooled methanol aqueous solution was used to extract the buds endogenous hormonesIAA, ABA, GA3and Z from chestnut, and the result of extraction was better. Throughexperimental analysis,The purifying method using solid phase extraction, SPE-C18cartridges was adopted, and methanol solution was chosen as the cleaning solution by thepreferred method, the elution solution: methanol/0.075%glacial acetic acid (90/5V/V),recovery ranged from65.7%to89.6%. Column: Symmetry300TM C18(5μm,4.6mm*250mm), gradient elution, the mobile phase: A was methanol, B was0.075%glacial aceticacid solution, flow rate:1.0mL/min; column temperature:30°C, injector Cover:10μL,detector: waters2996PDA. The samples were completely separated within25min, therelative standard deviation of IAA, ABA, GA3and Z were less than10%and therecovery ranged from65.7%to89.6%. The method was proved to be accurate, sensitiveand reproducible.2. The relationship of changes of the endogenous hormone content in chestnut during buddifferentiation stageThe IAA content has been showing a rising state during the process of flower buddifferentiation in chestnut (male inflorescence primordia formation),IAA content remainedat a high level, and ABA content increased firstly and then decreased, a slight increasefollowed fially; The GA3content increased at first and then decreased. It maintained at alow content state at last. the Z content increased firstly and then slowed down in theprocess,The relationship of Endogenous Hormone changes in the four kinds of varieties ofcultivar chestnuts were comprehensively analysed, chestnut bud tip before winter requireda certain concentration of IAA, IAA have larger impact on bud stage to the process;The low level of ABA was beneficial to differentiation and growth of chestnut flowercluster primordia; GA3was helpful to break dormancy and It was favorable to conversingto the inflorescence primordium and inflorescence primordium. Primordium and the flowerclusters required to maintain certain levels of GA3, and high levels of Z may have apromoting effect on the chestnut bud. In the analysis process of Z/IAA, GA3/IAA, ABA/IAA and GA3/Z value, Z/IAA, GA3/IAA and ABA/IAA showed a step change. It was aprocess of first increase and decrease followly, while GA3/Z value had been declining,and then maintained a lower value. The proportion of endogenous hormone in the chestnutbud in the entire process was analysed, high levels of Z/IAA, GA3/IAA and ABA/IAAwere conducive to chestnut shoots before winter breaking into flower bud dormancy period,but the period of the inflorescence primordia into flower clusters primordia required alower level of Z/IAA, GA3/IAA and ABA/IAA。Proportion of endogenous hormonesthrough trend analysis shows that chestnut flower bud tip before winter starts and thevarious stages of the process with the endogenous hormone balance are closely linked.3. Fertilization on endogenous hormone content of chestnutBy adjusting the N, P, K, Ca, Mg and B elements such as reasonable collocation,determined the chestnut is best time to fertilize, and fertilization. Studies show that all fivekinds of fertilization scheme can better promote the growth of tree.Fill the appropriate offertilization treatment input during June and July, the Chinese chestnut branch numberincreased obviously. The research results show that all test points in Chinese chestnutflower bud of IAA, ABA, GA3and Z content are higher than without fertilization.Applying boron fertilizer in March (fertilization scheme3) effects of several endogenoushormone levels are differ from that of other several schemes: IAA content in each test pointare highest, ABA content in inflorescence primordium differentiation stage (October toNovember) is higher than other fertilization scheme, in the flowers from primordiumdifferentiation stage (December to January) rise is higher than other period, the contents ofGA3and Z change with other fertilization scheme difference is not obvious. Early enoughB elements, accumulation of plant endogenous hormones also play an important role.Comprehensive analysis think, the choice of Chinese chestnut in March on N, P, K, Ca, compost, later (June to July) for B and N, P, K, Ca, which is mix ratio of fertilizer, canpromote the growth of Chinese chestnut. |
