Anoectochilus is one of the important Chinese herbal medicine. It mainly contains flavonoids, polysaccharide, trace elements, alkaloids, volatile oil, starch, vitamins and so on. According to the modern pharmacological research, Anoectochilus had the effects of hepatoprotective, anti-HBV, inhibition of LDL oxidation, hypoglycemic, hypotensive and bacteriostatic. There are mainly two kinds of Anoectochilus on the current market, Anoectochilus roxburghii (Wall)Lindl and Anoectochilus formosanus Hayata. This research focused on the effects of cultivating conditions and medium to the proliferation, growth and synthesis of main active ingredient of Anoectochilus, providing certain basic theory for production of high quality Anoectochilus and main active substance.According to the research which involved the influence of different culture conditions on the Anoectochilus roxburghii proliferation and the result showed that,the most suitable proliferation medium was MS+6-BA3.0mg/L+KT1.0mg/L+NAA 0.3mg/L+banana50g/L, with the proliferation rate of 3.83(P<0.0./).The most effective light for Anoectochilus roxburghii (Wall)Lindl proliferation was yellow light, and its multiplication coefficient reached up to 3.8 (P<0.01).According to the research which involved the influence of different culture conditions on the Anoectochilus biomass and the result showed that,the most suitable rooting medium for Anoectochilus roxburghii was 1/2MS+IBA 0.5 mg/L+KT 0.5 mg/L+potato 100 g/L, and its average fresh plant weighted 1.19g after four months’culture (P<0.01).The red light both promoted the growth of Anoectochilus roxburghii and Anoectochilus formosanus, and the average fresh plant weight groeed tol.33g and 1.47g respectively, which was significantly higher than control group (P<0.01).It researched the influence of different lighting conditions on the Anoectochilus’main active ingredient and the result showed that,the blue light which is the most effective light that induced synthesis of flavonoids, quercetin and isorhamnetin of Anoectochilus roxburghii, and the contents were 5.70 mg/g,54.07μg/g,27.15μg/g respectively, which was significantly higher than control group’s (P<0.01). While the yellow light which is the most effective light that induced synthesis of flavonoids, quercetin and isorhamnetin of Anoectochilus formosanus Hayata,and the contents were 10.69 mg/g,16.23μg/g,10.35μg/g respectively, which was also significantly higher than control group’s (P<0.01). when the photoperiod was 14h/d,and the contents of flavonoids, quercetin and isorhamnetin of Anoectochilus formosanus Hayata reached up to maximum,and the contents was 7.17 mg/g、6.32μg/g、6.81μg/g respectively.It researched the main active ingredient content between different Anoectochilus and different culture methods, and the result showed that, the total polysaccharide content of water culture seedlings and land culture seedlings’were both higher than the wild seedlings’. But the contents of total flavonoids, quercetin, isorhamnetin and kaempferol showed that,wild seedlings’> land culture seedlings’> tissue culture seedlings’> water culture seedlings’.The polysaccharide contents of different Anoectochilus showed that,Anoectochilus formosanus Hayata’s was higher than Anoectochilus roxburghii (Wall)Lindl’s from Sanming,and Anoectochilus roxburghii (Wall)Lindl’s from Zhangzhou was the lowest The contents of total flavonoids and flavonoid aglycones showed that, Anoectochilus roxburghii (Wall)Lindl’s from Sanming was higher than Anoectochilus formosanus Hayata’s, and Anoectochilus roxburghii (Wall) Lindl’s from Zhangzhou was also the lowest.
