Soil salinization is increasingly serious, which has become important limiting factor for soybean yield. Manganese acts many physiological functions in plant because of it’s different polyvalencies. In and out our country study indicates that exogenous manganese could enhance crop’s stress resistance. This study was conducted with soybean variety HN65 which is used in daqing area, and salt stress was simulated by sand culture and NaCl. Different foliage spraying treatments with EDTA-Mn,MnSO4, and MnCl2 were set to study the indexes related photosynthesis, stress-resistance physiological, metabonomics and proteomics, so as to clear the exogenous manganeseto improve the effect ofsoybean salt tolerance ability. The results were shown that:Compared with the control, foliage spraying with manganese could increase the content of chlorophyll, photosynthetic rate, Intercellular CO2 concentration, stomatal conductance, and MnSO4 and MnCl2 improved significantly some indexes related to photosynthesis under mild salt stress condition, and MnSO4 improved significantly under serious salt stress condition.Compared with the control, foliage spraying with manganese had increase the activities of protective enzymes, promote the ability to scavenge active oxygen and decrease significantly the content of H2O2, O2- and MDA, and increase the osmotic adjustment ability because of high content of proline, protein, soluble sugar.Metabolic profiling analysis indicated that foliage spraying with EDTA-Mn, MnSO4, and MnCl2 under mild salt stress condition induced 28, 17, 24 key different metabolites respectively, which belong to amino acid, organic acid, carbohydrate and sugar alcohols, phenolic acids, sterol and participate in secondary metabolism, ABC transporter, galactose metabolism, amino acid, biosynthesis of polyunsaturated fatty acids.Foliage spraying with manganese could increase the content of some metabolites increase and so improve the ability of plant osmotic regulation and anti-oxidation stress, conducive to the stability of cell.Proteomic analysis indicated that t foliage spraying with MnSO4 induced 342 different proteins, 235 ones were up-regulated, 107 ones were down-regulated. 31 paths of metabolism were changed, and 13 paths of metabolism including response to abiotic stimulus, oxidation reduction, biosynthesis of nitrogenous compounds, carboxylic acid biosynthesis, biosynthesis of organic acids, amine biosynthesis, amino acid biosynthesis, response to temperature stimulus, response to oxidative stress, biosynthesis of jasmonic acid, biosynthesis of amino acid derivatives, response to salt stress, glucose metabolism.
