Arprinocid was the only nucleoside species of anticoccidial drugs, with no cross-resistance for other coccidiostats, and having good anticoccidial effect to a variety of chicken coccidiosis. Abroad, arprinocid was added to feed for the prevention of coccidiosis, but currently our country had no raw material and any preparation of arprinocid approved. Our laboratory have developed a new formulation of arprinocid-arprinocid solution, intended to be administrated orally through drinking water for controlling coccidiosis in broilers, which was more consistent with clinical needs. To provide reference for rational application clinically, this issue carried out the study on the pharmacokinetics and pharmacodynamics of arprinocid solution in broilers. Details were as follows:1. The pharmacokinetics of arprinocid solution in broilersTo study the pharmacokinetics of arprinocid in broilers after orally taking the arprinocid solution, eight broilers were orally administrated with arprinocid solution at a single dose (14 mg-kg-1-bw). Blood samples were collected at different intervals and the arprinocid concentrations were detected by RP-HPLC. Then concentration-time dates were calculated with 3p97 program software.The result indicated that after orally taking the arprinocid solution, Arprinocid in chicken body fitted to one-compartment open model. The main pharmacokinetics parameters were in broilers as follows:Ti/2ka (0.69±0.57) h, Ti/2ke (5.02±2.27) h, Tpeak (1.91±0.83) h, Cmax(0.52±0.19) μg·mL-1, AUC (4.72±1.15) mg·h·L-1, CL/F (3.15±0.75) L-kg-1h-1, V/F (22.23±9.08) L·kg-1. The results showed that after oral administration of arprinocid solution, the arprinocid was absorbed rapidly and eliminated slowly in broilers.2. Efficacy of arprinocid solution against artifical infection of eimeria tenella in broilersThe effect of arprinocid solution against artificial infection of eimeria tenella was tested in broilers, to obtain the recommended dosage of arprinocid solution.240 broilers were randomly divided into eight groups, including five arprinocid solution groups of different levels (2,6,10,14,18 mg-kg-1·bw), toltrazuril treatment group(7 mg·kg-1·bw), healthy control group and infected-unmedicated group, and all the drugs were administrated orally through drinking water; the main efficacy evaluation indexes included bloody stool scores> the relative rate of weight gain, survival rate, cecal lesion and oocyst value, through which ACI was calculated, and the results of the anticoccidial drug efficacy were determined. The results showed that the ACI of the 2,6,10,14,18 mg·kg-1·bw of arprinocid solution and toltrazuril group were 103.7,143.5,170.5,195.3,193.2 and 195.9, respectively, indicating that 14 and 18 mg·kg-1·bw of arprinocid solution were highly effective anticoccidal drugs, and the former is better, comparable to toltrazuril. The recommended dosage of arprinocid solution against eimeria tenella in broilers was 14 mg·kg-1·bw .
