With the development of the economy in China and the rapid increasing number of pets,the number of skin defect case caused by burns,scald or car accident also increased gradually;The skin defect is mainly treated by methods of transplantation with autologous skin;In the transplantation with autologous skin,the survival of skin graft depended on the blood vessel regeneration of the host bed,so it demanded a thorough haemostasis and a rigid fixation of skin graft;FG had a role in fixation and good haemostasis effect.Thus in this experiment,FG was applied to the method of canine transplantation with autologous skin,with the purpose of assessing the influence on the transplantation and exploring preliminarily the mechanism of action of healing of skin graft with the applied of FG from molecular biology and histopathology,also provide theory on the applied of FG in the method of canine transplantation with autologous mesh skin.The canine skin defect model of forelimb were built at the first place of the experiment,whereafter compared study on the transplantation effect between the method of transplantation with autologous mesh and full-thickness skin were done to filter a better method of transplantation with autologous skins which showed high survival rate of autologous skins for subsequent experiments.FG was used in the better method afterwards,compared with the only use of the better method.According to the canine general condition,the local and histopathology,as well as the level of four kinds of cell factor of the autologous skins by immunohistochemistry on 3,5,7,14 and 28 d post operation,and the result showed that: 1.Compared with the method of transplantation with autologous full-thickness skin,the method of transplantation with autologous mesh skin showed a longer time of skin in vitro and a more difficult process technology,but more unobstructed drainage,slighter inflammatory reaction,higher survival rate and better effect of autologous skin. 2.Compared with the control group,canine of the experimental group acted less pain,and the local skin graft performed less errhysis and slighter inflammatory reaction but more obviously proliferation of fibroblast and new blood vessels,all above indicated that FG could enhance the healing speed of the skin graft and decrease the formation of scar,while the healing skin graft showed closer anatomical structure and physiological function to normal skin,finally promote the healing quality of skin graft. 3. The positive rate of b FGF: On 3,5,7and14 d post operation,the positive rate of b FGF of both the experimental group and the control group were keeping raise, and the positive rate of b FGF of the experimental group showed significantly higher than that of the control group(P<0.01).But on 28 d, the positive rate of the experimental group(18.67±0.88)showed significantly lower than that of the control group(30.67±0.88)(P<0.01). The results indicated that FG could enhance the level of b FGF,promote the proliferation of target cells such as fibroblast and vascular endothelial cell, speed up the tissue repairing and improve the ability of healing at the site of trauma. 4.The positive rate of VEGF: On 3,5,7and14 d post operation,the positive rate of VEGF of both the experimental group and the control group were keeping raise, and the positive rate of VEGF of the experimental group showed significantly higher than that of the control group(P<0.01).But on 28 d, the positive rate of the experimental group(21.33±1.45)showed significantly lower than that of the control group(28.60±1.20)(P<0.01). It was obvious that FG could enhance the level of VEGF,promote the proliferation and migration of vascular endothelial cell,and induced the formation of new blood vessel,while the number of trauma repairing cells was increasing which speed up the process of wound healing,therefore caused the ultima result that the healing of skin was improved. 5.The positive rate of P63 and integrinβ1: On 3,5,7and14 d post operation,the positive rate of P63 and integrinβ1 of the experimental group kept increasing and elevated to the maximum( 36.00±1.73 and 23.67±1.20)on 14 d,then decreased to 32.33±0.88 and 19.33±1.20 on 28d; The positive rate of P63 and integrinβ1 of the control group was increasing during the whole time and elevated to maximum on 28d(26.33±0.33 and 17.33±0.88);The positive rate of gene product p63 of the experimental group performed significant higher than that of the control group(P<0.01) on every time point,while the positive rate of integrin beta1 were significantly higher than that of the control of group on 5d, 7d and 14d(P<0.01).The results indicated that FG could enhance the expression level of P63 and integrinβ1 and promote the rapid proliferation of wound repairing cell such as epidermal stem cell,vascular endothelial cell,macrophage and fibroblast at the site of trauma,while the time of inflammatory reaction performed shorter,so the ultima result was that the skin healing was speeding.
