| Broomcorn Millet(Panicum miliaceum L.) also known as millet,as early as 10,300 years ago,it was cultivated domesticated as a main source in our country, Broomcorn Millet mainly grown in arid regions, commonly used in the salted ground improvement,desert governance and wasteland reclamation,while it can be use as a relief crop because of its short growth period. In addition,Broomcorn Millet also has a high nutritional value and health function,So far,few studies have reported about the germplasm resources of Broomcorn Millet at or out of abroad. In this study,we analysis the major bred vareties and their parents genetic diversity of broomcorn millet in domestic which uses our studies developed SSR markers. At the same time,created genetic populations that taking advantage of varieties bred and wild material,Combining screened SSR markers,constructed the molecular genetic map of broomcorn millet. The mainly findings are as follows:1)The genetic diversity of 88 accessions of broomcorn millet collected from various provinces in China was analyzed. The amplified fragments from 67 SSR primers exhibited moderate level of diversity in the broomcorn millet accessions investigated. In total,179 alleles were detected with an average of 2.7 alleles per locus. The polymorphism information content(PIC) and expected heterozygosity(He) ranged from 0.043 to 0.729(mean=0.376) and from 0.045 to 0.771(mean=0.445),respectively.2)The UPGMA clustering analysis at the genetic similarity value of 0.633 grouped the 88 accessions into four groups. Genetic structure assay indicated that there have close correlation between geographical regions and genetic diversity.This information is valuable for defining gene pools and developing breeding programs for broomcorn millet. Furthermore,millet-specific SSR markers in the study should serve an useful tools for assessment of genetic diversity and understanding of population structure in broomcorn millet.3)Conclusions The main results of the study are as follows: A total of 3506 pairs of polymorphic SSR primers screened, get more state 187 pairs of primers will get more state primer 150 pairs in the group after group of these primers in which one group How states have 130 pairs in the two groups in how states have 101 pairs withJoinmap3.0 primer data analysis software, groups can be connected on a 67 pairs of primers can be connected on two groups of 51 pairs4)Use the software win QTLcart to get Broomcorn Millet linkage map, which covers a population of 66.3c M, containing 13 linkage groups, the minimum distance between the marks on linkage group is 0.2c M, the maximum distance between the marks is 44.7c M,the average of the two markers distance is 63.2. Including at most 20 markers,at least 2 markers,contains an average of 4 markers on the linkage group. The longest linkage group covering about 126.7c M and the shortest of linkage groups covering only 24.6c M,the average length of each linkage group is 50.8c M. The group two covering 251.3c M,contains three linkage groups,the minimum distance between markers on linkage group is 0.7c M,the maximum distance between the marks is 46.5c M,the average distance between the two marks is 83.7c M. Including at most 23 marks,at least two markers,an average of 9 markers on linkage group. The longest linkage group covering about 160.3c M and the shortest of linkage groups covering only 46.5c M,the average of each linkage group is 9.3c M. |
