Like any other species, fish also need lots of nutrients that provide energy for their life activities, such as growth and development. In the process of evolution, fish differentiated into many kinds of feeding fishes( carnivorous, phytophagous, filter feeders, omnivorous), which all play an important role in aqueous system. Molecular studies on the shaping of fish’s food feeding and the differences of energy channel, which is bebefit for understanding the founction of fish. This study was divided into two parts, the first part is analysis of EST isozyme and –AMY isozyme in different fishes. The second part is about cloning and analyzing of partial LPL gene in different fish.First part, by using poyacrylamid gel electrophoresis, two isozymes(EST, α-AMY)in liver, muscle, heart, eye, gill, kidney of six kind of adult fish(Megalobrama terminalis、 Hemiculter leucisculus、 Cirrhina Cuvier、 Ctenopharyngodon idellus、 Mylopharyngodon piceus、 Siniperca chuatsi)was studied, We found nineteen kinds of EST isozymes and five kinds of AMY isozymes. The results showed that : the EST and α-AMY isozymes were very complicated and all the isozyme phenotypes exhibited tissues-specificity. The EST were found in this tissues and have obvious tissue-specific. The species and express degree of the EST isozyme was higher than α-AMY isozyme in these fish. The activities of the EST isozyme and α-AMY isozym in liver and kidney were higher than those in other organs. There exited difference in different feeding fish, so analysis on the characteristic of EST and α-AMY isozyme could reveal the current status of feeding habitat, knowing the nutrition source and ecological condition of fish habitats.Second part, is about the partial sequence of grass carp LPL gene. 1058 bp, gene was gotten by cloned with PCR. By analyzing of it, we can concude that, this sequence contained exon I(68 bp), exon II(169 bp), intron I(821 bp). Exon part compared with the m RNA sequence in grass carp LPL gene(FJ716100.1)from NCBI database, the similarity is 99.58%.The system of evolutionary tree reflects the evolution of LPL gene and kinship relations. According to the results of the evolutionary tree construted by partial LPL gene sequences of grass carp and partial LPL gene in 5 kinds of fish which searched in NCBI database, the results showed that grass carp and chub, culter alburnus and megalobrama, common carp and crucian, was respectively gathered for a class.
