The Sophora Viciifolia is the local species in karst area of Guizhou Province, which not only adapts to the soil conditions that drought, heat, partial acidic soil and barren soil in the karst areas,also has the feeding value such as abundant nutrition, more branches and leaves, good palatability and so on. And the medicinal value of roots, leaves, flowers and fruits of Sophora Viciifolia can improve the environment and achieve higher economic benefits, so the Sophora Viciifolia is an excellent species resources which is according with the sustainable development of Guizhou Province. Six kinds of Sophora Viciifolia groups from different altitudes were chosen to be materials in this study(the altitude was increasing from Group A to F), and researched on from three levels that morphology, seed storage protein and ISSR molecular marker and drew the following conclusions:(1) The morphological indexes of tested materials(Group E and F) from high altitudes were significantly lower than other 4 groups from lower altitudes(shoot height, stem diameter, the number of branch and branch length), the degree of variation of every morphological characters were similar; There existed variation among four morphological indexes of Sophora Viciifolia, and the variable coefficient of stem diameter was biggest.(2) There was hard seeds phenomenon of six Sophora Viciifolia populations in different degrees, the rate of hard seeds from Group A to Group F was 85%、92%、78%、85%、94%、95%respectively. According to analysis, the rate of hard seeds of Group E and Group F in high altitude were highest, which were significantly higher than others; The germination rate, germination force and germination index of Group C were higher than other populations and showed greater seed vigor.(3) There were differences among the degree of the number of spectral bands, position,width and shade in storage protein patterns of six Sophora Viciifolia populations in Guizhou Province,which can be used for making fingerprint.(4) Analyzing the genetic diversity of six different Sophora Viciifolia populations in Guizhou Province by two markers that seed storage protein and ISSR, the results showed that the genetic variation of Sophora Viciifolia mainly existed in the population, but the genetic differentiation level was higher between populations.(5) Comprehended two methods of ISSR molecular markers and seed storage protein markers of six Sophora Viciifolia groups in Guizhou, which showed the relationships of genetic diversity levels was Group F >Group D >Group E >Group C >Group A >Group B.(6) Clustering results showed that genetic distance among Sophora Viciifolia groups were bigger with the altitudes increasing, and the genetic distance of ophora Viciifolia groups that were adjacent altitudes was closer.
