Through determination and analyze of glandularia form, density and excretion during five tobacco varieties of Nanjiang 3, Honghua Dajinyuan, K326, Yun 87 and Yun 97, this research found out the restricting factors of glandularia excretion content in Nanjiang 3, and then enhanced the terpenoid metabolic precursor, IPP, content through DXPS and DXR gene overexpression in Nanjiang 3. This could improve the content of monoterpene, diterpenoid,polyterpene and their ramification in Nanjiang 3 in total, which could improve the glandularia excretion content in Nanjiang 3. The results show as follow:1. As all of them contained the long-handle and short-handle glandularia, only bifurcation glandularia in K326, there were not too much differences of glandularia form between five tobacco varieties. The order of total density between five varieties showed:Nanjiang 3> K326> Honghua Dajinyuan> Yun 87>Yun 97. Between different varieties, the density of long-handle glandularia was bigger than short-handle, which showed a reducing trend from upper to lower leaf, and decreased with the mature of tobacco.2. Between five varieties, in mature, the order of petroleum ether extract from high to low showed: Yun 97> Honghua Dajinyuan>Yun 87>K326>Nanjiang 3. According to the correlation analyze between petroleum ether extract and glandularia excretion, the content of petroleum ether extract was not in proportion to glandularia excretion.3. The order of glandularia excretion content from high to low showed: Yun 87>Yun 97>Honghua Dajinyuan> K326>Nanjiang 3. In Yun 87 and Yun 97, the detection was deduced as the main reason of high glandularia excretion, of which terpene alcohol, fatty alcohol and decomposition product of westpac alkanes. These materials are all main product of decomposition of monoterpene and diterpenoid, which was lower in Nanjiang 3 and Honghua Dajinyuan.4. Between five varieties, petroleum ether extract content was high as high glandularia excretion content, which could deduce that there was a certain correlation but not proportionbetween petroleum ether extract and glandularia excretion. Therefore, the glandularia excretion content could be increased by increasing compound of terpenoid, which could accomplish the increasing of tobacco oil content.5. This research built the plant expression vector p SH737-DXPS-DXR and identified 41 transgenic tobaccos by PCR after Nanjiang 3’s genetic transformation. In transgenic line DR6,there were significant change on leaf ingredient and increasing of main ingredient in extract.Directly detection of neophytadiene and topped alcohol showed that it is possible to enhance the terpenoid in Nanjiang 3 through DXPS and DXR gene overexpression.
