Apple(Malus×domestica Borkh) is one of the main species of fruit cultivation, its area and production rank first in the world.The development of apple industry, and to increase farmers’ income, adjusting agricultural structure is of great importance.But in actual production, in the course of cultivation of apple producing areas of part of shandong, shaanxi, henan, because the two season summer, autumn rain is more prone to flooding season, in a lot of rain, improper irrigation and drainage of bad factors, make the orchard soil under flooded conditions, often lead to apple fruit tree leaf etiolation, fall off, fruit quality and yield decline, serious when they make the whole plant death, cause serious economic losses. This has become in current apple production a problem to be solved. Therefore, it is necessary for the genetic mechanism of waterlogging resistance of apple trees were studied, to provide of apple root stock theoretical basis for revealing the waterlogging injury mechanism and adaptability mechanism.In this study G41(Ottawa 3×Robusta 5) and Sieversii(M sieverii(Ledeb.) Roem) for parents, F1 hybrid groups and parents as material, Among them G41 waterlogging resistance、resistance to disease and dwarf,Sieversii such as don’t resistant to waterlogging and drought resistance、qiaohua traits, The development of new SSR and SNP of markers, fine positioning apple stock waterlogging resistance genes.The main results are as follows:(1) Make analysis of variance and statistical test for the trait of F1 apple stock groups with 95% confidence interval, the trait including plant height, pitch number, internodal length and divarication number.The results showed that the plant height, pitch number, internodal length and divarication number of F1 apple stock groups were significantly different(P < 0.01).(2) The waterlogging tolerance of G41 x Malus sieversii F1 groups showed significantly different, the 5th day, Part of the strain began to suffer symptoms, after flooding 18 days, Nearly 30% of strain showed no suffer symptoms, but nearly 70% of the strains showed different suffer symptoms, Main Expressions were tip or leaf turn yellow, leaves fall off when the problem gets more serious,, finally led to the death of plants.(3) Make high-throughput sequencing of parents and two extreme mixing pool(DNA) with SLAF upgrade technology to parents and two extreme mixing pool(resistance to waterlogging DNA), identify the polymorphism of both parents, mainly SNPS, In Del. Put these tags in two extreme mixing pool for verification, to identify DNA markers whith showed polymorphism in the two extreme mixing pool, these tags was the mark of target gene chain.Through the functional annotation, genotype and phenotype correlation analysis, to ascertain the related flood-resistant gene loci of apple stock groups.
