With Suzhou garden was listed as world cultural heritage, its visibility co nstantly improve. Some well-known Suzhou garden had thousands of people ever y day in the busy season, sometimes reaching more than tens of thousands of pe ople. A large number of tourists got into the garden day in and day out inevit ably lead to the destruction of the garden.Over exploitation of tourism resource s, led to the emergence of various problems in the protection of gardens, the ec ological environment of gardens had also been some damage. How to take touris m resources of garden as stainable development was a big problem of protection of Suzhou gardens. Therefore, the study on ecological environment of Suzhou w as crucial.As the saying goes: ‘No water, no garden’,water was an indispensable land scape of Suzhou gardens, and plants directly reflected the greening effect of gardens. So in the previous studies, the researchers were in the sights of plants and water of Suzhou garden.However, sediment and soil as the matrix of both was little research. The domestic and foreignresearch reports were not found on t he microbial garden environment. This is the first of Suzhou garden in sediment and soil microbial diversity and community structure. There was no research rep orts about the environmental microbiology of garden found in domestic and forei gn 。 This work is the first study of Suzhou garden of microbial diversity and com munity structure in sediment and soil.Sediment and soil of several typical Suzhou gardens were collected for analysis, and analyze its physicchemical properties.According to the principle of metagenomics, using the methods of molecular biology widely used in the study of environmental microbial diversity--terminal restriction fragment length polymorphism(T-RFLP) technology analysis microbial diversity and community structure of sediment and soil.Moisture content,pH value, electrical conductivity,total organic compoun ds(TOC)and total nitrogen(TN)five physicochemical indexes of sediment an d soil in several Suzhou gardens were in the normal range. High content of t he total nitrogen brings hidden trouble to eutrophication. There are obvious diffe rences of bacterial and fungal diversity in different habitats of Humble Administ rator’s Garden in sediment and soil, and bacterial diversity was significantly higher than that of fungi. There were dominant species in sediment bacteria and soil bacteria and fungi. There was small kinds of fungal species in sediment, and no obvious dominant species. Microbial diversity of sediment and soil was the highest in autumn, spring second, winter of the third, summer was the lowest.There were differences between bacterial diversity in sediment and soil of 5 ga rdens,and demonstrated a negative correlation between microbial diversity and the number of tourist.This study provided a basic data accumulation for pollution abatement of Suzhou gardens, had an important reference value in analyzing community succession of Suzhou Garden, provided technical support and theoretical basis for microbial diversity and community structure of Suzhou gardens.
