This Paper has Drynaria roosii Nakaike as the material to using the method of plant tissue culture by orthogonal experimental design and statistical analysis methods, research the effects to spores in vitro propagation by the factor of explants sterilization time、sampling time of spore、basic medium、plant growth regulator、 prudential transplanting plant mass formula。Through these methods can be selected the suitable culture conditions for each stage of training. According to the research of in vitro culture technique on Drynaria roosii Nakaike which in order to establish Drynaria roosii Nakaike tissue culture and rapid propagation technology system. The results shows:1. Different sterilization time pollution problems affecting reproductive spores, Drynaria roosii Nakaike spores primary culture, The best sterilization effect:Through treating with 70% alcohol filter for 30 seconds、0.1% mercuric chloride for 60 seconds; The rate of obtaining sterile explants is 46.7%; The best basic culture medium is MS。2. Sterile culture the best sampling time is in late of May, because of the time when the spore mature best suitable as explants, and the spores are more mature, Sterilization time is also better grasp. The rate of spore germination is 80%, and growth in good condition, is conducive to sterile culture.3. Plant growth regulators on the Spore propagation subculture and proliferation has great influence,6-BA is the main influence factor of multiplication culture on Drynaria roosii Nakaike. The best hormone combination is MS+6-BA0.5mg/L+IBA0.1mg/L+NAA0.1mg/L, which is the most suitable of multiplication culture. Proliferation rate reached 1.56.4. The best medium of rooting culture is 1/4MS+0.2mg/L IB A, the rooting rate was 97%, the average number of roots was 4.6, the average root length was 3.2cm. Growth hormone is an essential component of rooting, activated carbon had no significant effect on rooting.5. Transplanting stage:Drynaria roosii Nakaike transplanted to loose and water retention is the best garden topsoil matrix, growth in good condition, the survival rate was up to 98%.
