| An experiment was carried out to proceed the gallus transcriptome biological assays and monitor the effects of Tanshinone IIA on AS. The first, 75 15-days Arbor Acre commercial broilers were divided into control group(20) and diseased group(55) randomly,the control group was drunk tap water and the diseased group was given 0.3% Nacl water from the 15 th day. The birds were killed when they harmed by the AS, the tissues of organs was collected and fixed in formalin for production of pathological sections, and the pulmonary arterys were collected for RNA-seq test. The second, 200 15-days Arbor Acre commercial broilers were divided into 4 groups: control, prevention, cure and diseased group. All except control group(tap water) were feed with Nacl water(0.3%) from 15th;the broilers in prevention group(0.02%) and treatment group(0.04%) were feed a diet with supplementary TIIA from 15 th and 29 th day respectively. Ten chickens were weighted and killed each group at day 28 th, 35 th, 42 nd and 49th; the right to total ventricular(RV:TV) and related biochemical indicators were determined. Results: compared to the control group,the broilers in diseased group grow slowly and have lots of pathological changes in organs and 895 differential expressed genes, the AS could disrupt 1990 GO Terms, such as Ribosome,Translation, and 122 KEGG metabolic pathway such as Ribosome,Cytokine-cytokine receptor interaction. Results showed that the number of broilers whose RV/TV between 0.22 and 0.25 in diseased group was significantly higher than the others. And the body weight in diseased group were significantly lower than in the other groups(P<0.01 or P<0.05). Compared to the control group, the concentration of plasma Tp, ALB, GLB,T-BIL, ALP, TG in the diseased group were significantly increased(P<0.01 or P<0.05), the concentrations in the cure group were significantly increased in the 28 th day( P<0.05),they in the prevention group changed no significant. The contents of the plasma BUN, CR,UA were significantly elevated(P<0.01 or P<0.05), they in the cure or prevention group were mostly no significant difference( P>0.05). The concentrations of Ca, P, K, Mg, Na,Cl in the diseased group were all significantly lifted(P<0.01 or P<0.05), the concentrations of Ca, Na, P in the prevention and cure group, the K, Mg, Cl in the cure group were lifted significantly also in the 28 th day( P<0.05), they in the cure or prevention group showed no significant difference in the 35 th to 49 th day. This study reveals the damage of Ascites syndrome to broilers on genes and metabolic pathway and et al., puts foward that the broilers whose RV/TV value between 0.22 and 0.25 are in a sub-health state, proves the Tanshinone IIA could prevent or cure the diseased in some extent, it provides some theoretical basis for the clinical prevention and cure of AS. |
