Hydatid disease is caused by Echinococcus multilocularis metacestode(larva) cause a zoonotic parasitic diseases, which hosts a wide range, but the sheep are the most suitable intermediate hosts, dogs are suitable for the end host. Hydatid disease prevention and control measures can inhibit terminals from adult Echinococcus eggs in the host to switch off the pathogen cycle chain, thereby reducing the spread of Hydatid disease. In the development of the parasite inside the intermediate host, the larval stage the protoscolex(protoscolex,PSC),the protoscolex in vitro conditions, but develops into a vesicle, for the control of echinococcosis in the molecular study, drug intervention, provided a good model.This study is the use of Echinococcus granulosus and development-related genes that have mature adult Egm123 be reprogrammed into the plasmids pET41 b and transfer to Escherichia coli, by inducing fusion protein expression and purification of GST-EgM123 and identification of immune activity of this protein, SDS-page showed expression of fusion protein molecule is about 56.1 kD. GST-EgM123 fusion to immunize New Zealand White rabbits, hyperimmune serum preparation and antigenicity of protein expression by Western blot identification, determination of antibody titer by ELISA method and its potency is above1:320,000.By protoscolex in vitro technologies detect the EgM123 antiserum on the rate of killing,adding fresh Guinea pig serum complement under specific antibodies on killing protoscolex by rate. Protoscolex GST-EgM123 hyperimmune serum, His-EgM123 hyperimmune serum and two kinds of high serum+Guinea pig serum-free cultured, the same time and the serum content of training conditions, hyperimmune serum with control group observation on protoscolices of Echinococcus granulosus in vitro immune effects. Results displayed, in high from serum group in the two species serum are out live fill body Hou, and control group compared to, original head Echinococcus granulosus mortality differences not significantly(P>0.05); normal serum group in 200 μL high from serum role 6 days, original head Echinococcus granulosus mortality respectively for 42.53% and 51.8%, and control group compared to, differences significantly(P<0.05); joined normal Guinea pig serum(containing fill body) of high from serum group, and control group compared to, original head Echinococcus granulosus mortality up 100%, differences significantly(P<0.05). These results indicated, fabricated by EgM123 antiserum protoscolex in vitro immune effects, lays the groundwork for development of Hydatid disease vaccines and related diagnostic.
