During this research, the following experiments were carried out:a completely randomized design of two factors, two different kinds of stock named shoot stock and root stock grafted with four different kinds of scion named shoot 1, shoot 2, shoot 3, shoot 4 from the morphology top of a branch, a completely randomized design of single factor, the different scion leaf area, half leaf,1 leaf,2 leaves, and 3 leaves and an orthogonal experiment of different plant growth regulators, different application methods, and dosages were carried out to study the Development of the grafted seedlings, the survival rate and the growth status of them.In the experiments, paraffin sections of each treatment was made, Motic BA 410 biology microscope and Moticas Pro 252A, Image Advanced 3.2 were together used to observe and measure the callus area of the grafted union; the soluble sugar, soluble protein content and POD activity of the Development were measured by physiological methods; the seedling height, diameter and seedling survival rate of different treatments were surveyed; Using PCA (Principal Component Analysis) to decrease the number of varieties without losing the main information of these 43 indexes basing on the seedling height, diameter and seedling survival rate of different treatments. Finally analyzed the Development of Camellia oleifera nurse seed graft and influencing factors.Results showed that:(1) Different treatments didn’t change the progress of the morphology changing, and the same Development is 5 stages:isolation layer, callus differentiation and induction, connection and of stock and scion and filling the graft union, cambium differentiation and induction, grooming tissue differentiation and induction. But different treatment changed the healing speed, the soluble sugar, soluble protein content and POD activity of the Development and the seedling height, diameter and seedling survival rate.(2) Chang Lin 4#,18#,23#,40# are different on healing speed, the soluble sugar, soluble protein content and POD activity of the Development and the seedling height, diameter and seedling survival rate, and Chang Lin 40# is the best among them,4# is better, then 23#,18# is the last one.(3) Shoot stock is better than root stock, basing on the fact that during Development, the healing rate of shoot stock grafted seedlings is 10% higher than root stock grafted seedlings, physiological index change curves of shoot stock grafted seedlings are always above those of root stock grafted seedlings, the seedling height, diameter and seedling survival of shoot stock grafted seedlings is 15.8%,33.9% and 36.3% higher than root stock grafted seedlings.(4) Within the shoot 1, shoot 2, shoot 3, shoot 4 numbered from the morphology top of a branch, shoot 2 is the best, and there is not too much difference between shoot 1, shoot 3 and shoot 4.(5) Among the different scion leaf area,1 leaf is best, half leaf is better, then 2 leaves and 3 leaves rank last. Data shows that the healing rate of 1 leaf is more than 90% after grafted 35 days, and the healing rate of half leaf is 90% after grafted 60 days, while the healing rate of 2 leaves, and 3 leaves approach 80% prospectively after grafted 60 days. Physiological index of different leaf area has no marked difference, the seedling height, diameter, seedling survival rate of 1 leaf is 90.2%,37.7%, and 31.7% higher than 2 leaves and 3 leaves, while the seedling height, diameter, seedling survival rate of half leaf is 47.8%,18.7% and 11.1%.(6) The best combination of plant growth regulator is:0.030 g·L-1 GGR6+0.2 g·L-1-1.0 g-L-1 IBA+0.14 ml·L"1-0.28 ml·L-1 Brassinolide+leaching medium.(7) Using PCA (Principal Component Analysis) to decrease the number of these 43 indexes and get 8 principal components:PC1 is the soluble protein of day 30, day 35 and day 40, PC2 is the soluble sugar of day 15, day 20 and day 25, PC3 is the healing rate of day 35, day 40 and day 45 and day 50, PC4 is the soluble sugar of day 35, day 40, day 50 and day 60, PC5 is the POD activity of day 5 and day 10, PC6 is the POD activity of day 35 and day 40, PC7 is the healing rate of day 5 and day 10, PC8 is the soluble protein of day 0 and day 5, these 8 principal components reflect 90.18% information of the origin varieties.
