Broussonetia papyrifera is one kind of deciduous trees, family Moraceae.Therefore, B. papyrifera has been used as raw material of high- grade papermaking and as sucrose-decreasing herbs;in addition, it could be used as raw material of feeds and plant species for barren mo untains afforesting. Currently, the major pathway to produce Broussonetia papyrifera seedlings was through seed channel and cutting channel. However, it’s difficult for seeds to germinate due to hard seed coat-caused moisture shortage, together with the properties of land occupancy, low efficiency, high cost, and limitation of seasons of cutting channel, it’s necessary to produce the seedlings in large scale with the aid of plant tissue culture.The explants was from the hybrid plants of B. papyrifera which was achieved by space breeding, and induction of callus, and embryonic cells, and somatic embryos was carried out, and then lots of vigorous somatic embryos were achieved. Here, the effects of phytohormones on induction of somatic embryos were investigated, and the main results are as follows:1.Young leaves of broussonetia papyrifera was cultured in 0.5mg/L MS medium containing 2,4-D for 7days, highest induction rate reach 93%. Tender leaves of broussonetia papyrifera was hardly cultured in MS medium whic h contained both of 2,4-D and 6-BA, but only get the bigger leaves, and tender broussonetia papyrifera leaves was browned obviously for 8 days;2. The pale yellow callus grew vigorously and soft, when was cultured in 0.2mg/L MS medium contained 2,4-D. When cultured in the light for 12h/d, callus could avoid lignification effectively. Below this concentration, easily appearing root differentiation with browning;3.Created the cell suspension culture system, obtain the highest induction rate when it was cultured in MS liquid medium with 1.5mg/L 6-BA and 0.5mg/L NAA;4. The highest rate of somatic embryogenesis was 17.9%, when hybrid broussonetia papyrifera callus was cultured in MS medium with 2.0mg/L 6-BA and 1.0mg/L NAA;5.The highest rate of artificial se eds germination was 33.3%, when sodium alginate was added to MS culture medium and 2.0mg/L 6-BA and 20g/L sucrose.
