Using stem segments as explants, this paper studied the antiseptic disinfection and differentiation, the optimal plant growth regulator combination and concentration in shoot proliferation and rooting, initially established high-quality variety paper mulberry regeneration system. Meanwhile, through the comparation of Salt Tolerance between hybrid paper-mulberry and wild paper-mulberry, we obtained the level of salt tolerance between them. The main contents and results were listed as follows:1. There is a big difference between different combination of different sterilizing agents and time.It shows that both the sterilizing agents and time should be considered to increasing the survival ratio. In this experiment the best combination is 3 and the survival could get 44%after 12 mins sterilization of 0.1%HgCl2.2. The stem segment were induced into adventitious buds; The most suitable medium for induction of the lateral bud was MS+6-BA1.0mg/L+NAA0.1mg/L, the induction rate was 94.4%, differentiation rate was 1.61,with a good growing state of green and tall.3. The concentration of NAA has a significant effect on the callus differentiation during the process of differentiation and growth, and the callus differentiation increased with the increase of NAA concentration. The concentration of 6-BA has the most effection on the stem's proliferation. Meanwhile, The concentration of GA has a significant effect on shoot's height. Taking all the above into consideration, MS supplemented with 6-BA and GA could improve culture multiplication and growth, MS+6-BA1.0 mg/L+GA0.2 mg/L is the best medium, and the average differentiation rate of adventitious bud is 3.3 with the average height of 2.83 cm.4. Considering the rootage time, rootage rate and the state of the root, the suitable medium for young plant grown and rootage at the same time was 1/2MS+NAA(0.3mg/L)+IBA(0.5mg/L) +6-BA(0.01mg/L) and 1/2MS+NAA(0.5mg/L)+IBA(0.1mg/L)+6-BA (O.Olmg/L), the rootage rate both reached at 100%.5. With the increasing of salinity concentration, the morphological changes of the different tree species after adding salt were different.The symptoms of the leaves could displayed the trend under salt stress with the stress time. The effects of salt stress on the chlorophyll contents of seedlings were various in different periods.The chlorophyll decomposed, the chlorophyll a,b and total chlorophyll contents were not regular, but the ratios of chlorophyll a to b increased or decrease. With the increasing of the salt concentration and time, the proline of the two plants rose differently, but the sugar and protein were not regular. The MDA had raised in the hybrid paper-mulberry, while had no obvious change in the wild paper-mulberry. It shows that there were different react between the two trees under salt stress.6. The comprehensive salt tolerant ability of wild paper-mulberry and hybrid paper-mulberry seedlings were appraised with subordinate function, the result showed the salt tolerant ability of wild paper-mulberry were stronger than hybrid paper-mulberry.
